摘要 目的研究腺病毒(adenovirus,Ad)12型E1B 55kD癌蛋白(ad12 E1B 55kD)与人Daxx(human Daxx,hDaxx)的相互作用及作用部位。方法利用细胞内外共免疫沉淀反应研究Ad12 E1B 55kD与bDaxx的直接结合反应,通过酵母双杂交体系测定两种蛋白质的相互作用。结果酵母双杂交试验结果显示Ad12 E1B 55kD通过全序列氨基酸(amino acid,aa)与hDaxx结合并发生相互作用。共免疫沉淀反应和Western blot结果证实Ad12 E1B 55KD能在细胞内外与hDaxx直接结合。结论Ad12 E1B55kD与hDaxx结合并发生相互作用。 Objective To question the reason of co-localization of hDaxx and PML in PODs disrupted by Adenovirus(Ad) type 12 E1B 55-kilodalton oncoprotein (Adl2 ElB 55kDa), the binding reaction and interaction of Adl2 ElB 55kDa with hDaxx was studied. Methods The interaction of Adl2 ElB 55kD with hDaxx was analyzed by the yeast two-hybrid assay. The binding of Adl2 ElB 55kD with hDaxx was studied by coimmunoprecipitation reaction in vivo or in vitro. Results Adl2 ElB 55kD oncoprotein interacted with hDaxx by binding the the full length amino acid(aa). Adl2 ElB 55kDa binds hDaxx in vivo or in vitro. Conclusion Adl2 ElB 55kD oncoprotein binds and interacts with hDaxx.
