摘要
目的:研究反义寡核苷酸(asON)体外抗汉坦病毒(HV)的效应。方法:设计合成了分别针对HVS基因片段5'端手柄状结构区的asON1和针对mRNA翻译起始区的asON2,采用双抗体夹心酶联免疫吸附试验(ELISA)检测asON作用后HV感染细胞培养上清中HV核蛋白(NP)的表达水平,免疫荧光试验(IFA)检测asON作用后感染细胞膜上NP的表达水平,空斑减少中和试验(PRNT)测定asON作用和HV感染细胞培养上清液中病毒滴度。结果:asON可抑制NP的合成以及病毒活性。结论:asON具有一定的抗HV作用,有可能作为一种治疗人HV感染的新途径。
Objective:To find out the antiviral effects of antisense oligodoxynucleotide(asON)in vitro.Methods:We designed and synthesized asON targeted at the Hantavirus76-118strain S gene initia-tion re gion of translation,5'terminal pan han dle region and noncomplementary sequence control.Their an-tiviral activity was evaluated in vitro by IFA,ELISA and PRNT.Results:asONs were able to effectively in hibit Hantavirus nucleo capsid pro tein expression and viral activity.Conclusion:The synthesized asON has,to some extent,antiviral ac tivity.As a new means,available for treating human Hantavirus infec tions.
