用MALDI_TOF MS观察适配子与炭疽芽孢的结合反应 被引量:1

摘要 用基质辅助激光解吸电离_飞行时间质谱(MALDI_TOFMS)观察炭疽芽孢适配子与炭疽芽孢结合反应 ,将SELEX(systemevolutionofligandsbyexponentialenrichment)技术筛选获得的一个炭疽芽孢适配子F77和第18轮的混合适配子以及兔抗炭疽芽孢抗体分别与炭疽芽孢结合反应后 ,用MALDI_TOFMS指纹图变化来研究这些分子与芽孢的结合情况 ,结果显示抗体、单个和混合适配子与芽孢结合反应后约在m/z1800处产生的质谱峰强度与芽孢对照相比明显降低;用MALDI_TOFMS质谱图变化可以推测适配子与芽孢的结合反应 ,该试验中观察到的结合情况与作者以前的研究结果一致。 Binding reactions of aptamers against spores of Bacillus anthracis with anthrax spores were investigated and evaluated by matrix assisted laser desorption ionization time-of-flight mass spectometry(MALDI-TOF MS).Aptamer F77,aptamer pool screened by system evolution of ligands by exponential enrichment(SELEX)technique at the18th round and rabbit antibodies to anthrax spores were reacted with anthrax spores,separately.After washing,the reactants were analyzed by linear MALDI-TOF MS.The peaks at about m/z1800of all the mass spectra were obviously lower than those of corresponding controls.MALDI-TOF MS could be used for detecting binding of aptamers with anthrax spores as a rapid and reliable analytical technique.The results obtained by MALDI-TOF MS were consistent with the previous conclusion published by the authors.
