摘要
目的 探讨脑缺血灌流后转录修复偶联因子ERCC6mRNA表达与神经元氧化损伤的关系。方法 采用局灶性脑缺血再灌流大鼠模型 ,按再灌注时间不同分 6组 ,用原位杂交法和免疫组化法分别检测ERCC6mRNA和 8-羟基 - 2′ -脱氧鸟苷 (8-OHdG)表达。结果 脑缺血再灌注 2 4h时 ,ERCC6mRNA杂交阳性细胞在缺血区的额、顶叶皮质、外侧纹状体散在分布 ,再灌注 48h缺血区的ERCC6mRNA杂交阳性细胞表达 ,72h时ERCC6mRNA表达达高峰 ,1 68h开始下降 ;8-OHdG阳性细胞数于2 4h时达高峰 ,48h时开始下降。结论 脑缺血再灌流后转录修复偶联因子表达增强 。
Objective To study the relationship between transcription-repair coupling factor ERCC6 expression and neuronal oxidative damage after cerebral ischemia and reperfusion.Methods By using Wistar rats' model of focal cerebral ischemia, in-situ hybridization and immunohistochemical method were used for showing ERCC 6 mRNA expression and neuronal DNA damage respectively.Results The number of ERCC 6 mRNA expression in the penumbra area increased at 48 h of reperfusion,reached peak values at 72 h,and decreased to a lower level at 168 h;8-OHdG positive cells reached peck at 24 h ,and started to decrease at 48 h.Conclusion The expression of ERCC 6 mRNA played an important role in relieving neuronal oxidative damage.
