人绒毛膜促性腺激素(hCG)抑制人PBMC促炎细胞因子mRNA的表达 被引量:3

摘要 目的 :探讨人绒毛膜促性腺激素 (hCG)对促炎细胞因子基因表达是否有影响。方法 :人PBMC与不同浓度的hCG(10 0、5 0、2 5、12 5、6 2 5、3 12 5U ml)共同于 37℃、5 %CO2 条件下培养 4小时 ,培养细胞行RT PCR测定 ,RT PCR产物经琼脂糖电泳后用图象分析仪作半定量测定 ,比较各组结果。结果 :5 0~ 6 2 5U ml的hCG对TNF αmRNA的表达有抑制作用 ,且与不加hCG的对照组比较有显著差异 (P <0 0 1) ;而 2 5及 12 5U ml的hCG对PBMCIL 1及IL 6mRNA的表达均有明显抑制作用(P <0 0 1)。结论 :hCG在一定剂量范围内有抑制促炎细胞因子mRNA表达的作用 ,提示在正常人体中也可存在的hCG在机体促炎 抗炎自稳机制中可能参与一定作用。 Objective:To investigate the effect of hCG on the gene expression of cytokines(TNFα?IL 1 and IL 6) that initiate inflammatory reaction protected against bacteria.Methods:Human PBMC were cultured with LPS(22 μg/ml) and different doses of hCG(100,50,25,12 5,6 25,3 125 U/ml) at 37℃,5%CO 2 for 4 h,mRNA of cells harvested were isolated,then run RT PCR with primer of TNFα?IL 1 and IL 6 separately.The PCR product were quantitated by using MIAS 2000 analyzer,calculated the OD value.Results:Compared with the control group,the result showed that hCG in a dose range of 50~6.25 U/ml have significantly suppressive effect on TNFα mRNA expression ( P <0 01),as well as on both IL 1 and IL 6 mRNA expression in a hCG dose range of 25~12.5 U/ml( P <0.01).There were no difference in the groups in which the hCG dose was higher or lower than that the dose in range above.Conclusion:It is suggested that hCG may involve in regulation of the production of cytokines that initiate inflammatory reaction against bacteria and may also explain part of the role of the hCG existence in normal human and cancer patients.
