摘要
目的调查多个实验室测定尿蛋白的方法以及结果,以便评价PRM法的性能。方法五份不同蛋白含量的尿液样品,由我省20家大医院检验科同时测定尿蛋白浓度;以判断PRM法的精密度、线性、准确度和检测下限等。结果不同的尿蛋白测定方法,其结果一致性较差;PRM法的日间精密度为1.31%(431.6mg/L)和1.12%(1916.4mg/L),尿蛋白含量在1530mg/L内线性良好,检测下限为62.5mg/L,平均回收率103.5%。测定结果与沉淀双缩脲法较一致,而磺柳酸比浊法测定值仅为该法的50%~87%。结论PRM法具有较好的可靠性和实用性,各实验室统一采用一种方法则具有意义且具可行性。
Objective To investigate methods and results of total urinary protein by analysed quantitatirely, and then to evaluate pyrogallol red-molybdate(PRM)method. Methods Five urine amples, whose protein concentrations are various, were measured by large twenty different clinical laboratories in our province, We experimented on in order to determine the precision, linearity, accuracy and sensitivity of PRM. Results There are different in results various methods. Within-run CV of PRM was 1.31%(431.6 mg/L)and 1.12%(1916.4 mg/L). The linearity was good if the urine protein levet doilt exceed 1530 mg/L, measuring low limit was 62.5 mg/L average recovery rate was 103.5%. Correlation was good between PRM and trichloroacetic acid-biuret method. The ratio of results of SSA quantitation and PRM quantitation was form 0.50 to 0.87; Conclusion Credibility and practicality of PRM is preferable, it is significant and feasible that all laboratories should abopt uniform method to determine total urinary protein.
