摘要
目的 将丹参与维拉帕米联合使用初步研究其对大鼠肝纤维化、肝硬化及门脉高压的作用效果 ,并探讨其机制。方法 采用高浓度CCl4 及低浓度乙醇诱导的肝硬化模型。于实验开始的第 4周、第 8周各组分别随机提取 6只动物 ,测定门静脉压力、谷氨酸转氨酶 (ALT)、透明质酸 (HA)及Ⅳ型胶原 (Ⅳ -C)。肝组织常规石蜡切片 ,HE染色 ,光镜下分析。结果 丹参及维拉帕米有降低门静脉压 ,降低转氨酶的作用 ,对肝纤维化指标也有改善 ,联合用药效果更佳。病理学表明 :联用组肝组织变性坏死程度、脂变程度、肝纤维化及肝硬化程度均显著低于模型组 ,联用组较单用组为佳。结论 丹参与维拉帕米联用可以有效地保护CCl4 /乙醇损伤大鼠肝功能 ,减轻肝纤维化及肝硬化程度。丹参与维拉帕米联用 ,可以有效地降低CCl4
Objective : To explore the effects of theearly and long term adminstration of radix salviae miltiorrhizae(RSM) and verapanil(Ver) on liver cirrhosis induced byccl 4 in rats. Methods The cirrhoticmodel of Wistar rats was established by(CCl 4)/ethanol.At the fourth and eighth weeks, portal vein pressure(PVP) of 6 rats from each group were measured. Theparameters of serum ALT,hyaluronic acid(HA),types Ⅳ collagen(Ⅳ C) and histological staining of hepatic tissue were evaluated. Results RSM and Ver could decrease the level of PVP,ALT,HA, Ⅳ C, and two drugs had better effect than single drug. In histopathology, two drugs could reduce the hepatocyte degenerationand nectosis and delay the formation of pseudolobe. Conclusion RSM plus Ver significantly decrease the extent of hepatic fibrosis and significantly reducethe portal hypertension in rats with CCl 4/ethanol induced cirrhosis.
