摘要
目的 检测不同剂量的重组人肿瘤坏死因子α(rhTNF -α)对涎腺腺样囊性癌细胞凋亡的体内诱导。方法 利用人涎腺腺样囊性癌细胞株 (SACC - 83)建立裸鼠移植瘤动物模型 ,局部肿瘤内直接注射rhTNF -α,选用两个不同剂量 (1 0 0× 1 0 4 IU/kg和 1 0× 1 0 4 IU/kg) ,观察对肿瘤的抑制作用 ,并采用流式细胞术定量检测体内细胞凋亡及细胞周期的变化情况。结果 ①A实验组的抑瘤率为 52 .1 6 % ,B实验组的抑瘤率为 73 .0 1 %。②用药后三组瘤体体积的变化有显著性差异。③三组流式细胞周期的变化及凋亡率有显著性差异。结论 ①rhTNF -α可抑制裸鼠SACC移植瘤生长 ,使细胞增殖在G1 期发生阻滞。这种抑制主要通过细胞凋亡来实现的。②本文低剂量的rhTNF -α诱导凋亡的效果优于高剂量的rhTNF -α 。
Objective To study the apoptosis of salivary adenoid cystic carcinoma induced by the different dosages of recombined human tumor necrosis factor alpha (rhTNF-α) at animal model.Methods Nude mice model was established by using salivary adenoid cystic carcinoma cell line (SACC-83).RhTNF-α was injected directly into tumor by two different dosages.The inhibition of tumors was observed and detected by flow cytometer.Results ①The ratio of tumor inhibition:group A was 52.16%,group B was 73.01%.②There was significant difference in tumor volume among three groups.③There was significant difference in the percentage of apoptotic cells and the changes in cell cycle distribution among three groups.Conclusion ①RhTNF-α can suppress SACC transplanted tumors in nude mice by inducing apoptosis.Cells proliferation were arrested in G 1 phase.②In this study,low dosage of rhTNF-α can play better inhibition role in the tumor than high dosage of rhTNF-α.
