摘要
分析了PCR扩增产物中非特异性电泳带和弥散状背景产生的主要原因.提出了单引物PCR扩增的几种分子模式,其一为5'端带有引物的单链DNA分子在3'端发生折转、自身结合和延伸,并在3'端形成同一引物的互补序列,变性展开后即可成为单引物PCR扩增的模板分子.讨论了单引物PCR扩增技术在遗传学和医学等领域中应用的可能性.
The molecular models of PCR amplification by single primers were presented. One model consists of primer and template DNA integrating specifically at one site. The 3′OH of primer was extended by DNA polymerase, the 3′OH of single chain DNA with primer reverses and extends and forms complementary sequence of the same primer at the 3′OH terminal. After heat-denatured the DNA molecule becomes a single chain DNA molecule in 5′ terminal with the primer and 3′ terminal with compelmentary sequence of the same primer. This single chain DNA molecule serves will as the template of PCR amplification by single primers. Reversed location of single chain DNA 3′ terminal witch primer refers to the nucleotide sequence of genome DNA template. This new technology of PCR amplification by single primers could be promising in its application in studies of genetics and medicine.
