酵母菌质膜蛋白质的形态和结构的研究

摘要 用冰冻蚀刻电子显微术观察了酵母菌质膜断面上的蛋白质。由于运用小角度旋转投影和某些新技术,不仅能区别开不同蛋白质的形态,位置,而且能区别开它们的亚基数目和排列方式。经图象分析后,测得质膜点阵区的蛋白密度为5.5×(10)~3个/μm^2。进一步将用示踪标记方法去探讨膜上蛋白质的形态,结构,运动等与功能的关系。 We used different shadowing methods and some new techniques on the plasmic intramembrane particles of Backerr s Yeast and prepared the specimen of freeze-fracture replica. It was discovered that rotary shadowing with small angle is reliable way to get high resolution replicas. It is able to distinguish not only shaps and sizes of differ- ent kinds of proteins but also numbers and arrangements of their subunits. Some of them consist of six subunits the others are four et al.Further work will be centered on relationship between shaped structure and function of the proteins on the membrane by cytochemistry, biochemistry et al.
