摘要
紫外辐射能明显地增加红豆杉细胞的异质性 ,紫外诱变单细胞使其植板率下降为原来的 1/ 3左右 .平板克隆培养 6 0d ,挑取克隆连续转接 4次 ,淘汰大部分后共得 78个克隆系 ,其中克隆系C4 2 B 0 9的生长速率和紫杉醇含量都较高 .生长速率和紫杉醇含量分别为亲本的 1.13倍和 6 .2 5倍左右 。
The variation of Taxus cells can be highly improved by UV irradiation and make the plating efficiency of single cell cloning of Taxus become 1/3 of the parent. The clones are chosen after 60 days of plating culture and transplanted for 4 times. 78 clone lines are obtained through many selections, among which the growth rate and taxol yield are 1.13 times and 6.25 times of the parent respectively and this clone line remains steady after three generations′ growth.
