摘要
Poly U与Poly C是两种核糖核苷酸的均聚物。在核酸结构与功能的研究上是不可缺少的生化试剂,特别是近年来在对抗癌药物的制备中应用更为广泛。本文的合成方法基本上是参照C. Basilio and S. Ochoa以及黎高沃等的报导,并根据具体条件做了一些改进,得到了满意的结果。一、材料 1.用大肠杆菌提纯的多核苷酸磷酸化酶粗酶液,生物物理所生化厂制备。 2.5’-尿嘧啶核苷二磷酸钠盐,含量95%,上海生化所制备。
Poly U and poly C were condensed from uridine-5'-diphosphate and cytidine-5'-diphosphate respectively by catalysis of polynucleotide phosphorylase isolated from E. Coli. The polymerization ratio of poly U synthesis was 62%, the sedimentation constant was 3.4S. The polymerization ratio of poly C synthesis was 68%, The sedimentation constant was 4.1S.
