柴胡疏肝散联合他莫昔芬对大鼠乳腺增生的干预作用 被引量:14

摘要 目的:观察柴胡疏肝散联合他莫昔芬对大鼠乳腺增生的干预作用。方法:50只随机大鼠分为对照组,模型组,柴胡疏肝散(6.3 g·kg^(-1))组,他莫昔芬(1.67 mg·kg^(-1))组,柴胡疏肝散+他莫昔芬组。采用夹尾激怒法制备大鼠乳腺增生模型,灌胃给药30d。酶联免疫法测定血清雌二醇,孕酮和催乳素,免疫组化法检测乳腺组织中雌激素受体及孕激素受体的表达。结果:与模型组比较,柴胡疏肝散联合他莫昔芬组大鼠血清中催乳素[(837.223±34.163)μg·L-1vs(346.215±43.114)μg·L-1,P<0.05]、雌二醇[(15.713±1.231)ng·L-1vs(4.973±0.278)ng·L-1,P<0.05)]的含量有效降低,孕酮[(0.423±0.031)μg·L-1vs(1.235±0.096)μg·L-1,P<0.05)]的含量提高;乳腺组织中雌激素受体表达显著被抑制(7.462±0.534 vs 4.889±0.343,P<0.05),孕激素受体的表达显著增强(1.212±0.53 vs 2.232±0.415,P<0.05)。结论:柴胡疏肝散联合他莫昔芬对大鼠乳腺增生有明显干预作用。 Objective:To study the protective effect of Chaihu Shugan San(CHSGS) combined with Tamoxifen(TAM) on Mammary Gland Hyperplasia of rats.Methods:50 rats were randomized divided into 5 groups,namely nomal,model,CHSGS(1.67mg·kg^(-1)),TAM(1.8 mg·kg^(-1)) and CHSGS combined with TAM group.Rats' model was established by clipping the tails of rates to cause Mammary Gland Hyperplasia.The intragastric administration period was 30 day.The levels of serum Estradiol(E2),Progesterone(P) and prolactin(PRL) were detected with ELISA.Immunohistochemical S-P technique method was used to evaluate the expressions of estrogen receptor(ER) and progesterone receptor(PR) protein in glandular tissue.Results:Compared with that of the model group,the levels of E2(15.713 ± 1.231 vs 4.973 ± 0.278,P < 0.01) and PRL(837.223 ± 34.163 vs346.215 ± 43.114,P < 0.01) of the CHSGS combined with TAM group were significantly lower,while the level of P(0.423 ±0.031 vs 1.235 ± 0.096,P < 0.01) of that group was significantly higher.Besides,the expression of ER of the CHSGS combined with TAM group was significantly inhibited(7.462 ± 0.534 vs 4.889 ± 0.343,P < 0.01),while the expression of PR of the CHSGS combined with TAM group significantly increased(1.212 ± 0.53 vs 2.232 ± 0.415,P < 0.01).Conclusion:CHSGS combined with TAM has significant protective effect on Mammary Gland Hyperplasia of rats.
