在附加有2,4—D^(3mg)/1并将蔗糖用量提高至^(50g)/1以及2,4—D^(5mg)/1、精氨酸^(5mg)/1和蔗糖为^(25g)/1的两种MS培养基上,两个品种的甘蔗幼叶的切段,经4—21天的无菌培养后,有90%以上的外植体产生了愈伤组织。石蜡切片观察表明,愈伤组织是由维管束鞘薄壁细胞分裂而来。用扫描电镜对比观察了密实和松散易碎的两类愈伤组织,后者有大的胞间隙,前者则无。
The young leaf-segment of two cultivar of sugarcane were cultured on MS-medium supplemented with 3mg/l 2,4-D and the sucrose were increased to 50g/l,or supplemented with 5mg/l 2,4-D,5mg/l arginine and 25g/l sucrose,over 90% explants were produced callus,after 4-21 days incubation. According to the observations of the paraffin section indicated that the callus were originates from the parenchyma cells of the vascular bundle sheath.Using the scanning electron microscope JSM-25, to compare the two kinds of calli, that the friable callus has many great intercellular spaces,and the compact callus has not.
