摘要
为了解河南省不同地区的牛群是否存在牛肠道病毒(Bovine enterovirus,BEV)感染及其感染情况,以便为BEV感染的防控提供流行病学资料,试验应用吉林大学动物检验检疫教研室建立的检测BEV抗原的双抗体夹心ELISA方法对采自河南省不同地区牛场的363份粪便样品进行检测,并对部分检测为阳性的病料进行病毒的分离培养、电镜观察及免疫学鉴定。将分离得到的阳性病毒毒株接种MDBK细胞后进行BEV 5′UTR基因的RT-PCR扩增及序列测定。结果表明:牛群的BEV感染率高达21.49%,不同地区牛群均存在程度不同的BEV感染率(2.50%~46.67%)。对ELISA检测为阳性的部分粪便样品进行病毒的分离培养、电镜观察及免疫学鉴定,获得了两株BEV毒株(BEV-HeN-YR2、BEV-HeN-YR91)。分离毒株的5′UTR基因序列的RT-PCR扩增产物测序后比对分析,两株BEV毒株均为F种肠道病毒。说明河南省不同地区牛群存在不同程度的BEV感染,且感染的BEV主要为F种。
To understand whether Bovine enterovirus(BEV)infection was present and its infection situation in cattle in Henan Province,and to provide epidemiological data for the prevention and control of BEV infection,the isolated from positive pathogen ELISA method established by Animal Disease Inspection and Quarantine Office in Jilin University was used to detect BEV antigens in 363 faecal samples from cattle farms in different areas of Henan Province.The virus were isolated,cultured,observed by electron microscope,and identified by immunological method.After inoculating MDBK cells with isolated positive virus strains,RT-PCR amplification and sequence determination of BEV 5′UTR gene were performed.The results showed that the overall BEV infection rate of cattle was as high as 21.49%,and there were different levels of BEV infection rate(2.50%to 46.67%)in different areas.Two strains of BEV strain(BEV-HeN-YR2,BEV-HeN-YR91)were obtained by isolation,culture,electron microscopic observation and immunological identification of some faecal samples tested positive by ELISA.It was found that both BEV isolates were confirmed to be enterovirus F(EV-F)enteroviruses after sequencing the RT-PCR amplification products of the 5′UTR gene sequence of the isolates.It indicates that there are different degrees of BEV infection in cattle in different areas of Henan Province,and the BEV infection ed is caused mainly by EV-F species.
