WDR1在肝癌组织中的表达及其与长期预后的关系

Expression of WDR1 in Hepatocellular Carcinoma Tissue and Its Relation to the Long-Term Prognosis

摘要 目的:探究WD40重复包含蛋白1(WDR1)在原发性肝癌组织中的表达以及临床价值。方法:免疫组织化学法检测106例肝细胞癌(HCC)患者的肝癌组织标本中WDR1的表达情况,统计分析WDR1与病理资料的关系及其与HCC患者长期预后的关系。结果:较之于癌旁组织,WDR1在肝癌组织中为阳性高表达。HCC中WDR1的高低表达情况与甲胎蛋白(AFP)、肝硬化、微血管侵犯、Child-Pugh分级、TNM分期等临床病理特征存在显著的相关性(P<0.05),而和肿瘤大小、性别等其余临床病理特征的相关性不明显(P>0.05)。COX回归分析表明WDR1是HCC患者总体生存预后的独立影响因素(HR=1.764,95%CI:1.270-2.841,P<0.05),也是HCC患者无瘤生存预后的独立影响因素(HR=1.803,95%CI:1.328-2.755,P<0.05)。Kaplan-Meier生存分析结果显示WDR1高表达组的总体生存率和无瘤生存率均显著低于WDR1低表达组。结论:WDR1是HCC患者的独立预后危险因素,且WDR1高表达者预后差。 Objective:To explore the expression of WD Repeat Domain 1(WDR1)in hepatocellular carcinoma tissue and to analyze the long-term prognosis.Methods:A total of 106 hepatocellular carcinoma patients treated in the XiangYa Hospital from February 2004 to December 2011 were enrolled.The expression of WDR1 in HCC tissue was detected by immunohistochemistry,and the relationship between WDR1 with pathological data and the patients’ long-term prognosis was analyzed by statistical software.Results:Compared with that in adjacent noncancerous tissue,the level of WDR1 increased in HCC tissues.The expression level of WDR1 in HCC had a significant correlation with HCC patients’ clinical pathological feature,such as,AFP,cirrhosis of liver,microvascularinvasion,Child-Pugh grading,or TNM stage(all P<0.05).And the correlation withthe rest of the clinical pathological characteristics,such as tumor size,gender,showed no statistic significance(P>0.05).Cox’s proportional hazards regression analysis showed that WDR1 was an independent influencing factor of over-all survival(HR=1.764,95%CI:1.270-2.841,P<0.05),and also an independent influencing factor of disease-free survival(HR=1.803,95%CI:1.328-2.755,P<0.05).The result of Kaplan-Meier survival analysis showed that overall survival rate and disease-free survival rate of high WDR1 expression group were significantly lower than those of lower WDR1 expression group.Conclusion:WDR1 is independent prognostic risk factors for HCC patients,and high WDR1 expression indicates the poor prognosis.
