摘要
目的:研究β-雌二醇(E2)对去卵巢小鼠子宫内膜组织中Hes-1基因表达水平的影响。方法:将16只雌性昆明系(KM)小鼠随机平均分为卵巢切除生理盐水干预组(OVX+NS)和卵巢切除β-雌二醇干预组(OVX+E2);采用Trizol一步法抽提内膜组织总RNA,进行RT-PCR实验,扩增出Hes-1基因和内参基因(GAPDH)目标条带,采用QuantityOne-4.6.2凝胶分析软件对Hes-1基因与GAPDH基因的目标带进行半定量分析。结果:OVX+NS组和OVX+E2组小鼠子宫内膜组织中Hes-1基因mRNA的相对表达强度分别为:0.16±0.06和0.06±0.01,P<0.05,具有统计学差异。结论:高剂量的β-E2可能通过下调Hes-1基因的表达促进子宫内膜上皮细胞增生,Hes-1基因和ER活性状态之间的相互作用可能在子宫内膜疾病的发展机制中发挥了重要作用。
Objective:To study the influence ofβ-estradiol(β-E2)interference on the Hes-1 gene expression level in uterine tissue of the ovariectomized mice.Methods:Sixteen healthy female KM(Chinese Kun Ming)mice were randomly divided into two groups according the treatment of ovariectomy plus normal saline administration(OVX+NS)and ovariectomy plus beta-estradiol intervention(OVX + E2);Trizol one-step technique was used to extract the total RNA for determination.RT-PCR was used to detect the expression of Hes-1 and GAPDH after amplification of the previous genes.Results:The relative intensities of Hes-1 mRNA expression in the endometria of the OVX+NS and OVX+E2 groups were 0.16±0.06 and 0.06±0.01,respectively.The Hes-1 mRNA expression level was significantly down-regulated in group OVX+E2 than in the group of OVX+NS(P<0.05).Conclusion:These findings suggest that high doses ofβ-E2 may promote endometrial epithelial cell hyperplasia by down-regulating the expression of Hes-1 gene.Interaction between Hes-1 gene and the ER activity may play a significant role in the development of endometrial diseases.
