藏绵羊EGLN1基因表达、变异特征及其适应性分析

摘要 为揭示藏绵羊高原低氧的适应机制,选择与高原动物低氧适应相关的EGLN1基因为研究对象,分析绵羊群体中该基因的遗传变异特征,为进一步寻求分子育种提供依据。采用荧光定量PCR及PCR-SSCP技术检测了藏绵羊和湖羊不同组织EGLN1基因表达量及其intron6-intron7区的核苷酸变异特征。结果显示:EGLN1基因在2个绵羊群体各组织间均有表达且存在组织差异性。其中,湖羊各组织的表达量均高于藏绵羊,在心脏、肝脏和肾脏组织差异最大。在2个绵羊群体EGLN1基因intron6-intron7区检测到了6个SNPs、1个碱基的插入和1个碱基的缺失位点,2个群体间等位基因和基因型频率存在统计学差异(P <0. 05),藏绵羊优势等位基因B以及优势基因型AB的频率极显著高于湖羊(P <0. 01)。说明藏绵羊EGLN1基因部分外显子及内含子区遗传变异程度高,多态性丰富,选择潜力大,可作为藏绵羊适应高原低氧环境的候选基因。研究结果将为藏绵羊的遗传改良和种质创新提供基础数据。 In order to reveal the adaptation mechanism of plateau hypoxia in Tibetan sheep,the EGLN1 gene was selected as the research object,and the genetic variation characteristics of the gene in sheep population were analyzed for molecular breeding. The quantitative PCR and PCR-SSCP techniques were used to detect the EGLN1 gene expression in different tissues and the nucleotide variation in intron6-intron7 regions of sheep. The results showed that the EGLN1 gene was expressed in the tissues of the two sheep populations,and there was a difference in tissues. Among them,the expression of Tibetan sheep was lower than Hu sheep,and it was higher in heart,liver and kidney. Six SNPs,one insertion and one deletion were detected. There was a statistically significant difference in alleles and genotype frequencies between the two groups( P < 0. 05). The frequency of dominant allele B and the predominant genotypes AB were significantly higher in Tibetan sheep than that in Hu sheep( P < 0. 01).The exon and intron regions of EGLN1 gene in Tibetan sheep had high genetic variation,rich polymorphism and great potential for selection. It can be used as a candidate gene for Tibetan sheep to adapt the high altitude hypoxia. The results will provide the basic data for genetic improvement and germplasm innovation in Tibetan sheep.
