地塞米松治疗坐骨神经低热损伤的实验研究 被引量:1

摘要 目的分析大鼠坐骨神经在低热损伤时予以地塞米松干预后的微结构变化,探索地塞米松对低热损伤的干预作用。方法将12只雄性Wistar大鼠按照随机化原则分为热损伤组(6只)与地塞米松干预组(6只),热损伤组大鼠的右侧坐骨神经给予47℃的热刺激,持续5 min;给予地塞米松干预组大鼠的右侧坐骨神经相同热刺激5 min,刺激结束后立即给大鼠腹腔注射地塞米松注射液。同时,每只大鼠的左侧坐骨神经可作为实验对照。在损伤后24 h,分析大鼠坐骨神经内的神经、血管微结构的改变。结果在损伤后1 d时许多有髓纤维已经出现轴索退变,血管内皮细胞出现肿胀,导致神经内膜中的毛细血管管腔发生狭窄。而地塞米松干预后,有髓纤维轴索退变改善不明显,且仍旧存在内皮细胞肿胀、部分毛细血管狭窄。结论使用地塞米松干预大鼠坐骨神经的低热损伤,对神经内的毛细血管、有髓纤维病变并没有明显改善作用。 Objective To observe the microstructure of sciatic nerves thermal injury after the treatment of dexamethasone, and to discuss the protective effect of dexamethasone in thermal injury. Methods A total of 12 male Wistar rats were randomly divided into thermal injury group(n=6) and the intervention group(n=6). The right sciatic nerves in the thermal injury group received thermal stimulation at 47℃ for 5 min, and those in the intervention group received dexamethasone in addition. The left sciatic nerves were exposed, as the control. Bilateral sciatic nerves were harvested after 24 h. The microstructure was examined in the microscope. Results On 1stday after the injury, many myelinated fibers had axonal degeneration, and vascular endothelial cells became swollen, leading to narrowing of capillary lumen in the endoneurosis. However, after the intervention of dexamethasone, the improvement of axonal degeneration of myelinated fibers was not obvious, and there were still endothelial cell swelling and partial capillary stenosis. Conclusion After intervention, the damage of endoneurial capillary and myelinated fiber degeneration are similar to that before.
