摘要
目的 探讨电针在脑损伤时对脑细胞凋亡和分泌型磷脂酶A2 (secretoryphospholipaseA2 ,sPLA2 )及其相关介质的影响。方法 采用线栓法制成大脑中动脉缺血再灌注 (middlecerebralarteryischemiareperfusion ,MCA IR)模型 ,用HE染色和TUNEL法观察脑细胞形态学变化 ,用免疫组化法检测sPLA2 在脑细胞中的表达 ,用 [3H]标记大肠杆菌为底物以及用ELISA法分别检测血清中sPLA2 活性和TNF α、PGE2 水平。结果 在大鼠MCA IR脑损伤时 ,电针能明显降低脑细胞凋亡与坏死 ,同时也可显著降低血清中sPLA2 、TNF α、PGE2 水平及sPLA2 在脑细胞中的表达 ,与对照组比较 ,差异有显著性意义 (P <0 .0 1)。结论 电针对脑损伤有良好的保护作用 ,其机理在于抑制sPLA2 的活性、蛋白表达及相关介质水平。
ObjectiveTo explore the effect of electroacupuncture on cerebral apoptosis, secretory phospholipase A 2(sPLA 2)and related mediators. MethodsThread occlusion technique was used to make models of middle cerebral artery ischemia reperfusion (MCA IR),apoptosis was measured by TUNEL staining method, while the sPLA 2 activity was detected by use of E.coli membrane labelled by oleic acid.The level of TNF α and PGE 2 were detected by ELISA test. sPLA 2 protein expression was measured by immunohistochemistry. ResultsIn contrast to control group, electroacupuncture inhibited the cerebral apoptosis and necrosis,it also remarkably inhibited the expression level of sPLA 2,TNF α and PGE 2 in rats with ischemic cerebral injury. ConclusionElectroacupuncture has a good protective effect on MCA IR injury of rats, which may be attributed to the decrease of sPLA 2,TNF α,PGE 2 level and the expression of sPLA 2 protein.
