摘要 目的 :为探讨血管紧张素转化酶 (ACE)I/D多态性影响血清ACE活力水平的可能机制。方法 :采用紫外比色法 ,以马尿酰甘氨酰甘氨酸为底物测定ACE活力和动力学常数 ,以PCR检测I/D多态性。结果 :4 5 %的活力水平改变由I/D多态性决定 ,DD、ID、II三种基因型之间的Km值无明显差异 ,Vmax则因I/D而异 ,依次为DD >ID >II。结论 :血清ACE水平与其I/D基因多态性有关 ,该多态性没有影响到酶与底物HGG的亲活性。 Objective:To investigate the possible mechanism that ACE I/D plymorphism induces the different levels of ACE activity in the serum. Methods: PCR was used to detect ACE I/D polymorphism and Hippurylglysylglycine(HGG)was used as substrate to test the serum ACE activity and kinetics constants. Results: 45% of ACE activity variance were modulated by I/D polymorphism. The Michaelis-Menten constants of ACE were not different from DD, ID or Ⅱ(p>0.05). Conclusion: The ACE I/D polymorphism is closely associated with variance of its activity in the serum and isn't involved in the affinity between the active site and substrate HGG.
