摘要 目的 探讨拓扑异构酶Ⅱ (TopoⅡα)及E型依赖钙黏附蛋白 (E cad)在口腔鳞状细胞癌发生、发展中的作用及临床意义。方法 应用免疫组织化学SP法检测TopoⅡα及E cad蛋白在正常口腔黏膜、炎性增生组织、不典型增生组织及鳞状细胞癌组织中的表达。结果 TopoⅡα在正常口腔黏膜、炎性增生组织中均为低表达 ,其在不典型增生组中的高表达率与其在口腔鳞状细胞癌组中的高表达率有显著性差异。E cad在口腔正常黏膜、炎性增生中的阳性率与其在鳞状细胞癌中的阳性率有显著性差异 ;E cad在口腔鳞状细胞癌组织中的阳性率与肿瘤淋巴结转移有显著相关性。结论 TopoⅡα及E cad在口腔鳞状细胞癌的发生中起一定作用 ;检测两者蛋白表达状况 ,有助于判断口腔鳞癌患者的预后和指导临床治疗。 Objective To study the role and clinical significance of expressions of TopoⅡαand E-cad during the carcinogenesis and development of oral squamous cell carcinoma. Methods The protein expressions of TopoⅡαand E-cad were examined by immunohistochemistry S-P method in oral normal mucosa, inflammatory hyperplasia, dysplasia mucosa and squamous cell carcinoma. Results The low expression of TopoⅡαwas seen in all normal mucosa and inflammatory hyperplasia, no high expression was observed. There was significant difference between dysplasia mucosa and squamous cell carcinoma. The positivity rate of E-cad was 100%,83.3%, 57.1%, 49.0% in oral normal mucosa, inflammatory hyperplasia, dysplasia musosa and squamous cell carcinoma respectively. There was significant difference between oral normal mucosa, inflammatory hyperplasia and squamous cell carcinoma.The expression of E-cad was related to the lymph node metastasis in oral squamous cell carcinoma. Couclusions Our data indicate that the abnormal expressions of TopoⅡαand E-cad play the role during the carcinogenesis of oral squamous cell carcinoma. Comprehensive analysis of TopoⅡαand E-cad will assist in evaluating prognosis and indicating the clinical therapy.
