摘要
通过3亲本交配,将供体菌株携带外源调节基因的质粒转移到植病生防细菌成团肠杆菌(E.agglomerans)IC1270中,得到4个衍生菌株。几丁质酶活性测定表明,衍生菌株对几丁质的消解能力均显著高于亲本菌株,以携带gacA或rpoD衍生菌株的活性最高。离体抗真菌颉抗实验表明,携带gacA或rpoD衍生菌株比亲本菌株对核盘菌和立枯丝核菌的抑制效果显著增强,其它衍生菌株无明显提高。
Four E.agglomerans IC1270 derivatives carrying heterologous regulatory gene (gacA, rpoD, rpoS, or apdA) from Pseudomonas fluorescens were constructed by triparental mating. Measuring chitinase activity showed that the chitinolytic activity of all the four derivatives was enhanced in comparison with their parental strain. The in vitro antifungal activity of the derivatives carrying gacA or rpoD was also markedly increased when tested with Sclerotinia sclerotiorum or Rhizoctonia solani as targets. The results suggested heterologous regulatory genes gacA and rpoD possess strong positive regulatory effect on elevating the biocontrol potential of E.agglomerans IC1270.
