摘要
将虾脊兰的种子接种到添加了马铃薯提取液、复合维生素作为添加物的不同有机物的培养基上,研究不同蔗糖浓度下有机添加物的效应。结果显示,单独添加有机物的促进效应,在10g/L蔗糖浓度下比在20g/L蔗糖浓度下更利于虾脊兰种子的萌发和生长,但生长指数和对照比较差异不显著,说明维生素或马铃薯提取液单独加入培养基效果不明显。两种添加物一起加入,对种子萌发和成苗的促进效应明显高于单独添加物的效应,生长指数高于单一添加物的生长指数,且差异显著,与对照的差异也显著。在10g/L蔗糖浓度下,两者同时添加的促进效应最好。故虾脊兰种子胚培养的最适培养基为VW+15%椰子汁+5%香蕉+10%马铃薯汁+1粒/L复合维生素胶囊+1%蔗糖。
Embryos from seeds of Calanthe rubens were cultured on modified VW+150?mL/L coconut juice+50?g/L banana,potato extract and multivitamins which were supplementd as additives to medium containing different level of sugar for seed germination and seedling development.The result showed that the promoting effects of adding a single additive in volume containing 10?g/L sugar was higher than that containing 20?g/L sugar,but their growth indices were not evidently higher than that of contrast.The effects of mixed organic additives were superior to the effect of single organic additive.So the mostly effective growth enhancer for seed germination and seedlings development was the combination of potato extract and multivitamins under 10?g/L sugar.The best medium for seed germination and sequent development of Calanthe rubenus was medium of VW+15%coconut juice+5% banana+1%sugar+1 capsule/L multivitamins+10% potato extract.
