摘要 目的:研究“补血活血”要药当归的主要有效成分-当归多糖(APS)对胎儿胸腺细胞、脾细胞表达造血生长因子GM-CSF、IL-3的影响。方法:采用造血祖细胞体外培养、造血生长因子生物学活性检测、免疫细胞化学、核酸探针原位杂交等实验技术。结果:APS诱导的胸腺细胞、脾细胞培养上清液可促进早期造血祖细胞CFU-Mix增殖分化;APS诱导可促进胸腺细胞、脾细胞表达GM-CSF、IL-3蛋白及mRNA。结论:APS可促进淋巴细胞表达造血生长因子GM-CSF、IL-3,进而促进人早期造血细胞发生。 Objective: To explore the effect of Angelica polysaccharide (APS) on the expression of GM-CSF and IL-3 from fetal thymocytes (FTC) and splenocytes (FSC). Methods: Techniques of hematopoietic progenitor cell culture in vitro, biological assay of hematopoietic growth factor (HGF), immunocytochemistry and in situ hybridization were used. Results: The conditioned media of FTC and FSC induced by APS can markedly promote the proliferation and differentiation of early multipotential progenitor cell (CFU-Mix);the expression of GM-CSF,IL-3 protein in FTC and FSC induced by APS were much stronger than that of the control; expression level of GM-CSF and IL-3mRNA showed similar tendency. Conclusion: APS promotes the expression of GM-CSF\IL-3 of FTC andFSC , then proceeds to promote early human hematopoiesis.
