摘要
目的 观察犬脑干持续缺血后Tau含量及磷脂酶A2 (PLA2 )活性的动态变化 ,探讨脑干缺血时Tau的神经保护作用及PLA2 的激活情况。方法 建立犬脑干缺血模型 ,动态观察缺血后各时点脑干组织中Tau含量 (应用美国Beck man公司生产的 63 0 0黄金系列高效氨基酸分析仪测定 )及PLA2 活性 (微量酸滴定法 )的改变 ,同时观察神经元病理损害情况。结果 与假手术组相比 ,缺血 3 0min ,Tau含量显著增加 (P <0 0 5 ) ,PLA2 活性亦显著增高 (P <0 0 5 ) ,神经元轻度异常 ;缺血 3h ,Tau含量增加更显著 (P <0 0 1) ,PLA2 活性非常显著升高 (P <0 0 1) ,神经元呈中度缺血改变 ;缺血 6~ 12h ,Tau含量及PLA2 活性仍继续增高 (P <0 0 1) ,神经元呈重度缺血改变或死亡。结论 脑干持续缺血后Tau的含量及PLA2的活性不断增高 ;
Objective To observe the dynamic changes of the content and the activity of phospholipase A 2(PLA 2) after ischemia in canine brainstem and to explore the neuroprotection of taurin and the activation of PLA 2. Methods The ischemic brainstem models were established with dog.The content of taurin and the activity of PLA 2 were determined after different durations of ischemia in the brainstem with Gold series high performance aminoacid analyzer and trace acid titration respectively. Meanwhile,the pathological changes of neurons were observed with microscope. Results Compared with those of the pseudo operation group,the content of taurin and the activity of PLA 2 in the brainstem increased significantly 30 min after ischemia in the brainstem ( P <0.05) and mild damages to neurons were found. At 3 h post ischemia, the increase of the content of taurin and the activity of PLA 2 became more significant ( P <0.01) and neurons turned to be moderate insult. During 6~12 h post ischemia in the brainstem, the content of the taurin and the activity of PLA 2 in brainstem continued increasing ( P <0.01), neurons became seriously injured and more and more neurons were found lost. Conclusion The content of the taurin and the activity of PLA 2 increase continuously after sustained ischemia in the brainstem. The increase of the content of taurin may be of neuroprotection at the early stage of ischemia in the brainstem.
