摘要 目的 :探讨一步离子交换层析法提纯牛视网膜可溶性抗原 (S Ag)。 方法 :利用DEAESepharoseFF凝胶及自动层析系统一步层析分离提纯牛视网膜S Ag ,聚丙烯酰胺凝胶电泳确定分子量 ,等电聚焦电泳确定等电点。结果 :分析证实 ,S Ag分子量 5 2 0 0 0 ,等电点 6.2 0。利用提纯S Ag成功地诱发出实验性自身免疫性葡萄膜视网膜炎 (EAU)动物模型。结论 :经一步层析法提纯的S Ag 。 Objective: Our aim was to isolate and purify soluble antigen (S Ag) from bovine retina by one step ion exchange chromatography. Methods: S Ag was isolated from bovine retina by ion exchange chromatography with DEAE sepharose fast flow and automatic gradient mixer system. The isolated S Ag was analyzed with sodium dodecyl sulfate polyacrylamide gel electrophoresis and isoelectric focusing. Results: The molecular weight of the purified S Ag is 52 000, and its isoelectric point is 6.20. The purified S Ag successfully induced the model of experimental autoimmune uveoretinitis (EAU). Conclusion: S Ag purified by one step ion exchange chromatography can be used for induction of EAU model.
