摘要
用BSA (BulkedSegregantAnalysis)法获得了与核桃早实性状相关的RAPD标记OPB 0 890 0 。混合分离群体的混合样来自早实品种辽 1的自然授粉实生后代。用 2 2 0个随机引物扩增早实和晚实混合样 ,只有引物OPB 0 8(5’GTCCACACGG 3’)在早实混合样及其个体上扩增出了约 90 0bp的特异带OPB 0 890 0 ,而在晚实混合样及其个体上无此特异带。用OPB 0 8为引物 ,以 19个品种和 3个株系的DNA为模板进行扩增 ,特异带OPB 0 890 0 在 17个早实品种中的 12个个体上出现 ,而 5个晚实品种 (系 )上均无此带。
A random amplified polymorphic DNA(RAPD)marker linked to the precocious trait of walnut( Juglans regia L.),OPB 08 900 ,was identified with bulked segregant analysis(BSA).Two bulks of DNA samples were created from open pollinated Liao 1(an precocious cultivar).The bulks were screened using 220 random primers(OP Kit).OPB 08(5'GTCCA CACGG 3')generated a specific fragment with precocious individuals and without backward individuals.The specific fragment of approximately 900 base pairs(bp),was designated as OPB 08 900 .22 DNAs from 19 cultivars and 3 clones were used as templates for RAPD analysis with primer OPB 08 respectively.The specific band of OPB 08 900 was present in 12 out of 17 precocious cultivars and absent in 5 backward cultivars/clones.The RAPD marker OPB 08 900 is related with precocious trait in walnut and would be used to distinguish the precocious trait and backward trait in walnut.
