摘要
目的:探讨獐牙菜苦苷对糖尿病周围神经痛(DPN)大鼠血清炎性因子水平的影响。方法:30只SPF级大鼠随机分为空白组、DPN组及獐牙菜苦苷组,每组各10只。链脲佐菌素(STZ)构建DPN大鼠模型,造模后第7、14天,獐牙菜苦苷组大鼠给予腹腔注射30 mg/kg獐牙菜苦苷;空白组与DPN组分别腹腔注射30 m L/kg的生理盐水。造模后第14天结束实验,采用ELISA、RT-PCR及免疫印迹法检测大鼠血清TNF-α、IL-6、IL-8、IL-10及TGF-β水平。结果:DPN组促炎因子TNF-α、IL-6、IL-8水平高于空白组,抑炎因子IL-10、TGF-β低于空白组(P<0.05)。獐牙菜苦苷组促炎因子低于DPN组,抑炎因子高于DPN组(P<0.05)。结论:獐牙菜苦苷可有效纠正DPN大鼠血清炎性因子失衡,有望成为治疗DPN的新药物。
Objective:To investigate the effect of swertiamarin on inflammatory factor levels in rats with diabetic peripheral neuropathy(DPN).Methods:Thirty SPF rats were randomly divided into three groups:blank group,DPN group and swertiamarin group(n=10 each).DPN rat model was established by streptozotocin(STZ).On days 7 and 14 after modeling,the swertiamarin group was given30 mg/kg Swertia Swertianoside via intraperitoneal injection,while the blank group and the DPN group were intraperitoneal injected with 30 ml/kg saline.The treatment was ended on day 14 after modeling,then the levels of serum TNF-α,IL-6,IL-8,IL-10 and TGF-βwere detected by ELISA,RT-PCR and Western-blot.Results:The DPN group showed higher proinflammatory factor levels and lower anti-inflammatory factor levels compared with the blank group(P<0.05).The swertiamarin group showed lower pro-inflammatory factor levels and higher anti-inflammatory factor levels compared with the DPN group(P<0.05).Conclusion:Swertiamarin may effectively correct the imbalanced inflammatory factor levels in DPN and is expected to become a promising new drug for DPN treatment.
