摘要 目的研究系统性红斑狼疮(SLE)患者外周血CD3+CD56+自然杀伤T细胞(NKT)的表达,探讨其临床意义。方法用流式细胞仪检测60例SLE患者及30例健康体检者(对照组)外周血CD3+CD56+NKT细胞表达水平,评估CD3+CD56+NKT细胞表达与SLE活动指数(SLEDAI)和实验室指标的相关性。结果 SLE患者CD3+CD56+NKT细胞表达水平[(2.03±0.21)%]明显低于对照组[(4.28±0.32)%],差异有统计学意义(P<0.05);SLE组IL-4水平为(41.82±10.41)pg/mL,明显高于对照组[(16.89±7.63)pg/mL],差异有统计学意义(P<0.05);CD3+CD56+NKT细胞表达水平与SLEDAI评分、24h尿蛋白定量、抗双链DNA抗体(抗ds-DNA抗体)及白细胞介素4(IL-4)水平呈负相关(r=-0.601,P<0.05;r=-0.584,P<0.05;r=-0.661,P<0.05;r=-0.528,P<0.05);与补体C3呈正相关(r=0.613,P<0.05)。结论 SLE患者体内存在CD3+CD56+NKT细胞数量下降和功能缺陷,可能与SLE的发病有关。 Objective To investigate the alteration of CD3+CD56+ NKT cells number in patients with systemic lupus erythema-tosus(SLE) and to analyze the relationship between CD3+CD56+ NKT and disease activity .Methods Blood samples were obtained from 60 patients with SLE and 30 healthy control subjects(control group) .The level of CD3+CD56+ NKT cells was detect by flow eytometry .The relationships between the level of CD3+CD56+ NKT cells and some laboratory parameters were analyzed .Results The level of CD3+CD56+ NKT in control group was(4 .28 ± 0 .32)% ,and (2 .03 ± 0 .21)% in SLE patients ,the difference was sta-tistically significant(P<0 .05) .The level of IL-4 in SLE patients was(41 .82 ± 10 .41)pg/mL ,which was significantly higher than that in control group[(16 .89 ± 7 .63)pg/mL ,P< 0 .05] .Negative correlation was found between the level of CD3+ CD56+ NKT cells and SLEDAI ,proteinuria ,IL-4 as well as anti-dsDNA .However ,positive correlation was found between the number of CD3+CD56+ NKT cells and level of C3 .Conclusion The number of CD3+CD56+ NKT cells in SLE patients decreases significantly and has close relationship with disease activity .
