摘要
细菌光敏色素是一类含有胆绿素BV分子、对红光和远红光具有响应的色素-蛋白复合物。本文在采用Atto495分子对细菌光敏色素同源二聚体进行荧光标记的基础上,借助二维相关光谱手段分析了SDS对于结合到蛋白中的Atto495荧光发射谱的影响,发现除了非特异性结合,Atto495可以与Cys20以及其它区域的Cys结合。相关工作为后续分析光转换过程中的蛋白结构的变化奠定了基础。
Bacteriophytochrome is one biliverdin-containing proteins,which can make response to red light and far-red light.In this work,Agp1 mutants were labelled by Atto495 fluorescence probe,and then 2Dspectroscpic methods were used to study the spectral changes induced by SDS.It was found that Atto495 could be noncovalently combined to the Agp1-500 apoprotein that contained no Cys residues.Meanwhile,Atto495 molecules could target Cys20 and other Cys residues in other domains.These studies provided some fundamental data for the further studies on the structural changes during the photoconversion process.
