穿心莲内酯对人肝癌SMMC-7721细胞增殖和细胞周期的影响

Effect of andrographolide on cell proliferation and cell cycle of human liver cancer cells line SMMC-7721 in vitro

摘要 目的探讨穿心莲内酯(AD)对人肝癌SMMC-7721细胞增殖和细胞周期的影响。方法不同浓度AD作用于SMMC-7721细胞,培养24,48,72 h收获细胞。四甲基偶氮唑盐比色法(MTT)检测SMMC-7721细胞的增殖情况;流式细胞术(FCM)检测SMMC-7721细胞周期的变化。结果不同浓度AD干预SMMC-7721细胞,培养24 h后,细胞呈现明显的形态学改变。MTT检测结果显示:随着AD浓度的增加,SMMC-7721细胞光密度值(OD)逐渐减小,呈时间-剂量依赖性,与对照组比较差异有统计意义(P<0.05或P<0.01)。流式细胞检测结果显示:SMMC-7721细胞阻滞于G0/G1期,与对照组比较差异有高度统计意义(P<0.01)。结论 AD对人肝癌SMMC-7721细胞增殖具有抑制作用,能使SMMC-7721细胞阻滞于细胞周期的G0/G1期,提示AD抗肿瘤的作用机制与抑制肿瘤细胞增殖有关。 Objective To observe the biological effect of andrographolide( AD) on cell proliferation and cell cycle of human liver cancer cell line SMMC-7721. Methods The SMMC-7721 cells were cultured by AD with different concentrations for 24 hours,48 hours and 72 hours respectively. The inhibition of cell proliferation was observed by microculture tetrazolium test( MTT); the changes of cell cycle in SMMC-7721 were detected by flow cytometry( FCM). Results The cultured SMMC-7721 cells were intervened by AD with different concentrations for24 hours,showed typical morphological changes. MTT showed that AD inhibited the SMMC-7721 growth in timeand dose-dependent manner. The proliferation rate tended to decrease along with the increase of AD dose. Compared with control group,there was a significant difference( P<0.05 or P<0.01). FCM showed the cultured SMMC-7721 cells were treated by AD for 48 hours with different concentrations,they were blocked on G0/ G1 stage. Compared with control group,there was a significant difference( P< 0.01). Conclusion AD can inhibit the proliferation of SMMC-7721 and make the SMMC-7721 cell block at G0/ G1 stage in the cell cycle. Anti-tumor mechanism correlates with the inhibition of cell proliferation.
