摘要
目的 研究局部使用外源性胰岛素样生长因子 1(Insulin likegrowthfactor 1,IGF 1)对骨骼肌钝性打击伤后愈合速度、质量和对内源性IGF 1和IGF 2的基因转录的影响。方法 1%硫喷妥钠 (每 10 0g体重 0 .5ml)麻醉后 ,对大鼠右下肢腓肠肌内侧面中段实施钝性打击伤 ,而后在创伤局部实施皮下肌膜外注射药物 ,以此分为 :生理盐水组 (对照组 )和IGF 11.5 μg组 (对照组 ) ,每组均作以下分析 :mRNA分析 ;组织学分析 ;电镜超微结构分析。 结果 伤后实验组比对照组更早形成基底板层保护膜 ,更早更多地激活成肌细胞、生成肌丝、形成肌管、融合成肌纤维 ,愈合质量也较好 ,实验组内源性IGF 1及IGF 2的mRNA含量均较对照组提早升高。结论 局部注射外源性胰岛素样生长因子 1可以使内源性的IGF 1和IGF 2的基因转录提早 ;可以刺激成肌细胞增生 ,促进肌管形成 ,加速肌管融合成肌纤维 ,加速骨骼肌创伤后修复过程 。
Objective To investigate the effects of extrinsic insulin like growth factor 1(IGF 1)on the healing process of injured muscle. Methods Following the establishment of models of skeletal muscles contusion, the rats were divided into the IGF 1 injection group and the control group, then the data obtained were analyzed. Results The formation of basement membrane was earlier in the experimental group. In the experimental group, both the myoblasts, myofibrials and the myotubes grew faster and more than that in the control group, indicating that the quality of healing process was better than that in the control group. Conclusion Local application of IGF 1 may exert favourable effects on the healing process after skeletal muscular trauma.
