葡萄-藏红花间作系统下尖孢镰刀菌拮抗菌Cs12对藏红花根腐病的抑制作用

摘要 [目的]以藏红花单作和葡萄与藏红花间作土壤根系土为研究对象,分析微生物群落组成和差异表达菌群。[方法]对湖州现代农业示范园葡萄园区葡萄-藏红花间作和藏红花单作种植地块进行根系土壤采集。利用RoToR HDA菌落高通量筛选工作站构建细菌资源库。通过对间作菌群进行分离纯化以及平板对峙筛选出有益菌。通过分析藏红花的发病率来探究有益菌对藏红花根腐病的抑制作用。[结果]通过对间作菌群进行分离纯化以及平板对峙,分离出1种尖孢镰刀菌拮抗菌Cs12。球体接种试验表明Cs12使病情指数降低50%,对尖孢镰刀菌的防治效果达75%。[结论]分离出1种新发现的有益菌Cs12,这种菌具有较好地拮抗尖孢镰刀菌的作用。藏红花-葡萄间作系统不仅能够解决藏红花腐烂病问题,而且提高了土地资源利用率。这种生物防治方法对于防治其他植物的土传病害有一定的参考价值,有助于农业产业的可持续发展。 [Objective]To analyze the microbial community composition and differentially expressed flora in the root soil of saffron monocropped soil and grape and saffron intercropping soil.[Method]Root soil was collected in the grape garden of Huzhou Modern Agriculture Demonstration Garden in the plot of grape and saffron intercropping and saffron monocropping.We constructed the bacterial resource bank using the RoToR HDA colony high-throughput screening workstation.The beneficial bacteria were screened by isolation and purification of intercropping bacteria and plate confrontation.The incidence of saffron was analyzed to explore the inhibitory effect of beneficial bacteria on saffron root rot.[Result]Through the isolation and purification of intercropping bacteria and plate confrontation,we isolated an antagonistic antibacterial agent of Fusarium oxysporum Cs12.The inoculation experiment showed that Cs12 reduced the incidence index by 50%,and the control effect of CS12 against Fusarium oxysporum was 75%.[Conclusion]We have isolated a newly discovered beneficial bacterium Cs12,which has a good antagonistic effect against Fusarium oxysporum.The saffron-grape intercropping system can not only solve the problem of saffron rot,but also improve the utilization rate of land resources.This biological control method has certain reference value for the control of soil-borne diseases of other plants,and is helpful for the sustainable development of agricultural industry.
