球毛壳菌发酵滤液对松树蜂共生真菌的抑菌活性

Antibacterial Activity of Fermentation Filtrate from Chaetomium globosum against Symbiotic Fungi of Sirex noctilio

摘要 探究樟子松(Pinus sylvestris var.mongolica)内生真菌球毛壳菌(Chaetomium globosum)发酵滤液对重大入侵害虫松树蜂(Sirex noctilio)共生真菌网隙裂粉韧革菌(Amylostereum areolatum)的抑制作用,筛选抑菌能力较强的差异代谢产物,有效防治松树蜂。首先对球毛壳菌发酵滤液抑菌活性进行测定,然后从代谢组学角度对球毛壳菌中潜在的抑菌物质进行筛选。结果表明,球毛壳菌发酵滤液可以完全抑制松树蜂共生真菌A.areolatum的菌丝生长,且抑菌活性稳定,不受储存时间、紫外线和温度的影响。共筛选出球毛壳菌发酵滤液差异代谢产物213种,其中潜在的抑菌物质15种,包括异阿魏酸、香草酸、大麻二酚、甘氨酸、大黄素、丹皮酚、4-胍基丁酸、隐绿原酸、苯并呋喃、4-羟基苯甲酸、绿原酸、琥珀酸、4-甲氧基肉桂酸、龙葵素和二甲喹唑啉C。差异代谢产物的KEGG富集分析中ABC转运蛋白、柠檬酸循环(TCA循环)、缬氨酸、亮氨酸和异亮氨酸生物合成、酪氨酸代谢、苯丙氨酸代谢、氨基苯甲酸盐降解丁酸代谢和C_(5)支链二元酸代谢显著富集。研究表明,球毛壳菌发酵滤液能够抑制松树蜂共生真菌网隙裂粉韧革菌的菌丝生长,且抑菌活性稳定,并筛选得到球毛壳菌发酵滤液的潜在抑菌物质。 To investigate the inhibitory effect of the fermentation filtrate of the endophytic fungus Chaetomium globosum isolated from Pinus sylvestris var.mongolica on symbiotic fungus Amylostereum areolatum of the significant invasive pest Sirex noctilio,and to screen differential metabolites with high inhibitory capacity,aiming at effectively controlling S.noctilio.The antibacterial activity of C.globosum fermentation filtrate was determined first,and then,potentially antibacterial substances were screened using a metabolomics approach.The results showed that the fermentation filtrate of C.globosum completely inhibited the mycelial growth of A.areolatum,the symbiotic fungus of S.noctilio,with stable antibacterial activity unaffected by storage time,ultraviolet rays and temperature.A total of 213 metabolites were screened out in C.globosum fermentation filtrate,including 15 potentially antibacterial substances namely isoferulic acid,vanillic acid,cannabipiperidiethanone,glycitein,emodin,paeonol,4-guanidinobutanoic acid,cryptochlorogenic acid,benzofuran,3-caffeoylquinic acid,4-hydroxybenzoic acid,chlorogenic acid,succinic acid,4-methoxycinnamic acid,solasodine and dictyoquinazol C.KEGG enrichment analysis of differential metabolites showed significant enrichment in pathways including ABC transporters,citrate cycle(TCA cycle),valine,leucine and isoleucine biosynthesis,tyrosine metabolism,phenylalanine metabolism,aminobenzoate degradation,butanoate metabolism and C_(5)-branched dibasic acid metabolism.Studies have shown that the fermentation solution of C.globosum inhibits the mycelial growth of the symbiotic fungus A.areolatum of S.noctilio s,with stable inhibition activity,and the potential inhibition substances can be screened.
