桔梗根系响应元宝枫凋落叶的转录组分析

摘要 【目的】探究北京地区元宝枫凋落叶对桔梗药用活性成分积累的影响,通过转录组测序揭示桔梗根部活性成分合成的分子机制。【方法】利用Illumina对桔梗根部进行转录组测序,分析萜类、类黄酮和苯丙烷类次生代谢产物合成通路中的基因表达变化。【结果】共检测到15727个SSR位点。发现753个DEGs注释到GO数据库的三大分类中,314个DEGs注释到KEGG数据库的105个代谢通路中。与对照组相比,分别有17,7,26个DEGs参与萜类、类黄酮和苯丙烷类化合物的生物合成。【结论】萜类和苯丙烷合成途径的DEGs主要上调表达,类黄酮合成途径的DEGs普遍下调表达。推测萜类和苯丙烷类物质合成上升,类黄酮合成下降。获得了桔梗根部的全部转录组信息,并初步预测了桔梗萜类、类黄酮以及苯丙烷类合成可能的调控通路。 [Objective]The aim of this study was to explore the effect of Acer truncatum fallen leaves in Beijing on the accumulation of medicinal active ingredients of Platycodon grandiflorum and to reveal the molecular mechanism of active ingredient synthesis in P.grandiflorum roots through transcriptome sequencing.[Methods]We performed transcriptome sequencing using Illumina high throughput sequencing technology.Gene expression changes in the synthesis pathways of terpenes,flavonoids,and phenylpropanoid secondary metabolites were analyzed.[Results]A total of 15727 SSR loci were detected.753 differentially expressed genes(DEGs)were annotated into three major categories of the GO classification,and 314 DEGs were classified into 105 metabolic pathways in the KEGG classification.Compared with the control group,there were 17,7,and 26 DEGs involved in the biosynthesis of terpenes,flavonoids,and phenylpropanoids,respectively.[Conclusion]We found that DEGs in the terpenes and phenylpropanoid synthesis pathways were mainly up-regulated,while DEGs in the flavonoid synthesis pathway were generally down-regulated.It is speculated that the synthesis of terpenes and phenylpropanoids is increased,while the synthesis of flavonoids is decreased.We obtained the transcriptome information of P.grandiflorum roots and preliminarily predicted the regulatory pathways for the synthesis of terpenes,flavonoids,and phenylpropanoids in P.grandiflorum.
