摘要
目的研究芪蛭益肾胶囊对高糖诱导的人肾小管上皮细胞(HK-2)损伤的保护效应及机制。方法将HK-2细胞随机分为对照组(DMEM培养基)、模型组(DMEM培养基+60 mg·mL^(-1)葡萄糖溶液)、芪蛭益肾胶囊最佳给药剂量组(DMEM培养基+60 mg·mL^(-1)葡萄糖溶液+300μg·mL^(-1)芪蛭益肾胶囊冻干粉溶液)进行培养。采用CCK-8法检测HK-2细胞存活率,活性氧检测试剂盒检测ROS含量,流式细胞仪检测细胞凋亡和细胞周期,ATP检测试剂盒检测ATP的含量。结果与对照组相比,模型组细胞存活率受到明显抑制,差异有统计学意义(P<0.01),与模型组相比,各芪蛭益肾胶囊给药组细胞存活率均升高,差异有统计学意义(P<0.01);与对照组相比,模型组HK-2细胞的凋亡比例增加,细胞G_(0)/G_(1)期比例增加(P<0.0001),G_(2)/M期+S期比例减少(P<0.0001),ROS含量提高(P<0.0001),ATP含量下降(P<0.0001);与模型组相比,芪蛭益肾胶囊组细胞凋亡比例减少,G_(0)/G_(1)期比例减少(P<0.05),G_(2)/M期+S期比例增加(P<0.05),细胞中ROS含量降低(P<0.05),ATP含量上升(P<0.01)。结论芪蛭益肾胶囊干预后减轻了高糖对HK-2细胞存活率的抑制,增加了G_(2)/M期+S期比例,降低了ROS含量,使ATP水平上升,说明芪蛭益肾胶囊对高糖诱导的HK-2细胞损伤具有保护作用。
Objective To study the protective effect and mechanism of Qizhi Yishen Capsules on HK-2 cell injury induced by high glucose.Methods HK-2 cells were randomly divided into control group(DMEM medium),model group(DMEM medium+60 mg·mL^(-1) glucose solution)and optimal dose group of Qizhi Yishen Capsules(DMEM medium+60 mg·mL^(-1) glucose solution+300μg·mL^(-1) Qizhi Yishen Capsules lyophilized powder solution).The survival rate of HK-2 cells was detected by CCK-8 method,the content of ROS was detected by reactive oxygen species detection kit,the apoptosis and cell cycle were detected by flow cytometry,and the content of ATP was detected by ATP detection kit.Results Compared with the control group,the cell survival rate of the model group was significantly inhibited,with significant difference(P<0.01).Compared with the model group,the cell survival rate of each Qizhi Yishen Capsules group was increased,and there was a significant difference(P<0.01).Compared with the control group,the apoptosis rate of HK-2 cells in the model group was significantly increased,the proportion of G_(0)/G_(1) phase was increased(P<0.0001),the proportion of G_(2)/M phase+S phase was decreased(P<0.0001),the content of ROS was significantly increased(P<0.0001),and the content of ATP was significantly decreased(P<0.0001).Compared with the model group,the proportion of apoptosis in the Qizhi Yishen Capsules group was decreased,the proportion of G_(0)/G_(1) phase was decreased(P<0.05),the proportion of G_(2)/M phase+S phase was increased(P<0.05),the ROS content in the cells was decreased(P<0.05),and the ATP content was increased(P<0.01).Conclusion After the intervention of Qizhi Yishen Capsules,the inhibition of high glucose on the survival rate of HK-2 cells was reduced,the proportion of G_(2)/M phase+S phase was increased,the content of ROS was decreased,and the level of ATP was increased,indicating that Qizhi Yishen Capsules has a significant protective effect on HK-2 cell injury induced by high glucose.
