耳石标记中水体锶浓度对梭鱼代谢与免疫相关酶学指标的影响

Effect of seawater strontium concentration on enzymes involved in metabolism and immunity of Liza haematocheila during otolith strontium marking

摘要 为探讨耳石标记中水体锶离子(Sr^(2+))浓度对梭鱼(Liza haematocheila)代谢与免疫相关酶学指标的影响,设计一个对照组(C0)和两个试验组(S1和S2),其中C0组梭鱼在正常海水(Sr^(2+)浓度为7.9 mg·L^(-1))中进行培养;S1和S2组梭鱼培养所用海水Sr^(2+)质量浓度分别为50 mg·L^(-1)和200 mg·L^(-1)。在标记实验开始后0.5、1、3、6、9、12、24、48、72、96 h,以及标记实验完成后24 h(即120 h),分别测定梭鱼的代谢相关酶学指标(PK和LDH)和免疫相关酶学指标(SOD和NOS)。结果显示:1)和C0组相比,S1组梭鱼代谢相关酶学指标(PK和LDH)和免疫相关酶学指标(SOD和NOS)未发生显著性变化(P>0.05);2)实验期间,S2组梭鱼PK值显著高于C0组(P<0.05);S2组梭鱼PK值在标记初期呈明显的上升趋势,随后下降到一个相对稳定水平,其变化趋势与C0组(PK值在实验开始后直接下降到一个相对稳定水平)有所不同;S2组梭鱼PK值在1、3、6、9 h时均显著高于C0组和S1组(P<0.05);3)和C0组相比,S2组梭鱼LDH、SOD和NOS未发生显著性变化(P>0.05)。结果表明,Sr^(2+)质量浓度50 mg·L^(-1)的标记环境对于梭鱼能量代谢和免疫功能均没有明显影响,而Sr^(2+)质量浓度200 mg·L^(-1)的标记环境仅提高了标记初期梭鱼对能量需求以应对高Sr^(2+)环境的压力,但这种压力尚未引起梭鱼机体缺氧现象的发生,免疫功能也未受到明显影响。从本研究结果可知,梭鱼耳石高Sr^(2+)标记期间,养殖管理措施可与正常梭鱼养殖管理措施一致。 In the process of otolith strontium marking of Liza haematocheila,it is necessary to add strontium to the growing environment,which may affect the metabolism and immunity of L.haematocheila.In order to explore the effect of seawater strontium concentration on enzymes involved in metabolism and immunity of L.haematocheila during strontium marking on otolith,two marking treatments(S1 and S2)and one control treatment(C0)were designed.In C0,L.haematocheila were raised in normal seawater,the Sr^(2+)concentration was 7.9 mg·L^(-1).The seawater used in treatment S1 and S2 were normal seawater with SrCl_(2)·6H_(2)O added,and the mass concentration of Sr^(2+)in seawater was 50 mg·L^(-1) and 200 mg·L^(-1),respectively.A total of 330 healthy juveniles with a body length of(3.00±0.26)cm were selected and evenly distributed to treatments C0,S1 and S2.During the strontium marking period,each treatment were consisted of 11 cuboid glass aquariums containing 30 L water,and 10 juveniles were placed in each aquarium.After 96 h of strontium marking,the remaining juveniles in each treatment were separately transferred to an aquarium with natural sea water and continued to be cultured until the end of the experiment.At 0.5,1,3,6,9,12,24,48,72,96 h of marking experiment and 24 h after marking experiment(120 h),five fish were sampled and prepared for further analysis.Pyruvate kinase(PK)and lactate dehydrogenase(LDH)were used as metabolism indicators,and superoxide dismutases(SOD)and nitric oxide synthase(NOS)were used as immunity indicators.The results were as follows:1)In seawater with 50 mg·L^(-1) of Sr^(2+),there was no significant effect on the metabolism-related enzyme indexes(PK and LDH)and immunerelated enzyme indexes(SOD and NOS)of L.haematocheila.2)In seawater with 200 mg·L^(-1) of Sr^(2+),PK activities increased first,then decreased to a relatively stable level,which was different from that of the control.And PK activities of L.haematocheila at 1,3,6,9 h were significantly higher than those of C0 and S1 treatments.The m
