摘要
目的:探讨麦粒灸对抑郁大鼠行为学、海马区环磷腺苷效应原件结合蛋白(CREB)的影响及对突触功能和突触可塑性的调控作用,探究麦粒灸对抑郁大鼠的防治作用及机制。方法:将40只SD雄性大鼠随机分为空白组、模型组、麦粒灸组、氟西汀组,每组10只。孤养+慢性不可预见性应激(CUMS)21 d复制抑郁大鼠模型。造模第1~21天,麦粒灸组在造模前1 h给予麦粒灸“心俞”“肝俞”“脾俞”“肾俞”穴3壮预防治疗,1次/d。同期氟西汀组予氟西汀溶液[1 mg/mL,10 mL/(kg·d)]灌胃治疗。观察各组大鼠的体质量及行为学改变。21 d后,行为学检测后2 h内解剖大鼠取出海马组织。免疫细胞化学技术(IHC)检测海马组织脑源性神经营养因子(BDNF)、代谢型谷氨酸受体1(mGluR1)、GluR1阳性表达情况。RT-PCR法检测海马组织BDNF mRNA和CREB mRNA相对表达量。Western blotting法检测CREB、BDNF、CaMKⅡ蛋白相对表达量。结果:与空白组比较,模型组大鼠体质量、糖水偏好率及旷场实验垂直运动频率和水平移动距离均下降(P<0.01),强迫游泳实验静止时间明显延长(P<0.01);IHC结果显示模型组mGluR1阳性表达明显升高(P<0.01),BDNF、GluR1阳性表达明显减少(P<0.01);Western blotting结果显示模型组CaMKⅡ、CREB和BDNF蛋白表达均显著下降(P<0.05);RT-PCR结果显示模型组CREB mRNA和BDNF mRNA表达均减少(P<0.01)。与模型组比较,麦粒灸组及氟西汀组以上指标出现相反变化(P<0.05)。麦粒灸组GluR1阳性表达高于氟西汀组(P<0.05)。两组其余各参数比较,差异均无统计学意义(P>0.05)。结论:麦粒灸法可通过调节抑郁大鼠CaMKII/CREB/BDNF信号通路相关蛋白质的表达,减轻谷氨酸诱导的神经毒性,发挥抗抑郁作用。
Objective:To explore the effects of wheat grain moxibustion on the behavior of depressed rats,the effects of cAMP-response element binding protein(CREB)in the hippocampus,and the regulation of synaptic function and synaptic plasticity in the hippocampus,in order to investigate the preventive and therapeutic effects of wheat grain moxibustion on depressed rats and its mechanism.Methods:Totally 40 SD male rats were randomly divided into blank group,model group,wheat grain moxibustion group,and fluoxetine group,with 10 rats in each group.The rat model of depression was replicated by solitary rearing+chronic unpredictable mild stress(CUMS)for 21 days.From the 1st to the 21st day of the modeling period,the wheat grain moxibustion group intervened by wheat grain moxibustion at"Xinshu(BL 15)","Ganshu(BL 18)","Pishu(BL 20)"and"Shenshu(BL 23)"points 1 hour before modeling.Each point was continuously moxibustion 3 times for preventive treatment,once a day.Meanwhile,the fluoxetine group was given fluoxetine solution[1 mg/mL,10 mL/(kgod)]by intragastric administration.Body mass and behavioral changes were observed in each group.After 21 days,hippocampal tissues were dissected and removed from the rats within 2 hours following behavioral testing.Positive expressions of BDNF,mGluR1,and GluR1 were detected by IHC.RT-PCR was used to detect relative expression of BDNF mRNA and CREB mRNA.Western blotting was used to detect protein expressions of CREB,BDNF and CaMKⅡ.Results:Compared with the blank group,the body mass,sugar-water preference rate,vertical movement frequency,and horizontal moving distance in open field test of rats decreased in the model group(P<0.01),while the resting time of rats in the forced swimming experiment was significantly prolonged in model group(P<0.01).The IHC results showed that the positive expression of mGluR1 was significantly increased in model group(P<0.01),while the positive expression of BDNF and GluR1 was significantly reduced(P<0.01).Western blotting results showed that CaMKII,CREB and BDNF pr
