摘要
目的建立蒙药山柰配方颗粒指纹图谱及多成分含量测定方法,并结合化学计量学对不同批次的山柰配方颗粒进行质量评价。方法采用HPLC法,色谱柱为COSMOSIL 5C18-AR-Ⅱ(4.6 mm×250 mm,5μm),流动相为甲醇-0.1%乙酸水溶液,流速为1.0 mL·min^(-1),柱温40℃,检测波长250 nm。采用《中药色谱指纹图谱相似度评价系统(2012年版)》进行指纹图谱相似度评价,SPSS 25.0软件进行聚类分析(CA),SIMCA 14.1进行主成分分析(PCA)和正交偏最小二乘法判别分析(OPLS-DA)。结果15批山柰配方颗粒指纹图谱相似度均>0.90,共标定12个共有峰,并指认出5种成分,绿原酸、香草酸、阿魏酸、异鼠李素-3-O-β-D-葡萄糖苷和对甲氧基肉桂酸乙酯含量分别为0.074~0.088、0.023~0.031、0.017~0.033、0.102~0.132、31.014~40.729 mg·g^(-1)。CA、PCA、OPLS-DA结果显示,15批山柰配方颗粒可分为两类,5个已知成分中,绿原酸等3个成分VIP值>1。结论建立的山柰配方颗粒指纹图谱及多成分含量测定方法准确、稳定、重复性好,化学计量学方法能够较好地筛选出标志性成分,可为山柰配方颗粒质量标准提供参考。
Objective To establish a fingerprint and multi-component content determination method for Mongolian traditional medicine Kaempferia formula granules,and to evaluate their quality by chemometrics.Methods The chromatography was performed on COSMOSIL 5C18-AR-Ⅱ column(4.6 mm×250 mm,5μm)with mobile phase consisting of methanol-0.1% acetic acid water at the flow rate of 1.0 mL·min-1.The column temperature was 40℃ and the detection wavelength was 250 nm.The Fingerprint Similarity Evaluation System of Traditional Chinese Medicine Chromatographic Fingerprint(2012 Edition)was used to evaluate the fingerprint similarity,and SPSS 25.0 software was used for clustering analysis.SIMCA 14.1 was used in principal component analysis and orthogonal partial least squares discriminant analysis.Results The similarity of fingerprint of 15 batches of Kaempferia formula granules was greater than 0.90,a total of 12 common peaks were calibrated,and 5 components were identified.The content of chlorogenic acid,vanillic acid,ferulic acid,isorhamnein-3-O-β-D-glucoside and ethyl p-methoxycinnamate ranged from 0.074~0.088 mg·g^(-1),0.023~0.031 mg·g^(-1),0.017~0.033 mg·g^(-1),0.102~0.132 mg·g^(-1),and 31.014~40.729 mg·g^(-1).The above three analyses showed that 15 batches of Kaempferia formula granules were divided into 2 groups.Among the 5 known components,3 components such as chlorogenic acid had values greater than 1.Conclusion The established fingerprint and multi-component content determination method is accurate,stable and reproducible.The chemometrics method can screen out the iconic components well,providing reference for the quality standard control of Kaempferia formula granules.
