SNP分子标记大熊猫妊娠鉴定初探

摘要 为探索大熊猫Ailuropoda melanoleuca妊娠鉴定分子技术,采集4只(2只采样当年产仔/2只采样当年未产仔)雌性大熊猫常态及配种后血液样品,分别提取常态血液样品中的总DNA 4份,以及配种后血液样品中的游离DNA 5份(其中1只采样2次),配种后的采样时间从减食前26 d至减食后1 d。对所有DNA样本进行基因组重测序,并在60个SNP位点上进行基因分型对比(常态/配种后)。4只雌性大熊猫配种后样本SNP位点数据均检测出差异基因型,表明所有个体当年均成功受孕。2只大熊猫个体最终未产仔,其中1只于减食前26 d检测到差异基因型,但差异基因型在减食后1 d消失。研究结果验证了分子标记用于大熊猫妊娠鉴定的可行性,为今后相关技术的进步与完善提供数据参考,同时提示流产可能是大熊猫繁育中的一种普遍现象,管理上可注意多方面调节,以期提高大熊猫圈养种群的繁殖率。 To explore the pregnancy diagnostic technique of giant pandas(Ailuropoda melanoleuca),9 DNA samples from 4 female giant pandas(2 parturient and 2 non-parturient)were collected and extracted.Four samples were collected(one from each female)before mating and 5 circulating free DNA(cfDNA)samples were collected after mating(2 samples were collected from the same female).The sampling time ranged from 26 d before appetite loss to 1 d after appetite loss.Sixty single nucleotide polymorphism loci on the DNA were genotyped by genome re-sequencing.The differential genotypes in cfDNA,which are considered as the evidence of pregnancy,were detected in all the four giant pandas,indicating the successful fertilization of these individuals.Specifically,in one of the two non-parturient giant pandas,the differential genotypes were detected 26 d before appetite loss,but disappeared 1 d after appetite loss.These results validated the feasi-bility of using molecular markers for the pregnancy diagnostics of giant pandas and provides reference for the development of future technologies.These findings suggest that abortion may be a common phenomenon of giant pandas and that appro-priate adjustments of animal husbandry may be helpful to improve the reproductive rate of captive giant pandas.
