基于网络药理学探讨五味甘露治疗银屑病机制

摘要 为探讨蒙药五味甘露治疗银屑病的潜在作用机制,为后期进一步实验研究提供理论依据。利用NCBI数据库、Swiss ADME和Swiss Target Prediction等挖掘蒙药五味甘露(小白蒿、水柏枝、麻黄、刺柏叶、照山白)的有效成分和靶点基因;利用TTD、GeneCards等数据库获取疾病的作用靶点,根据药物与疾病的交集靶点绘制Venn图,通过交集靶点获得蛋白互作关系图;通过Metascape平台进行GO功能和KEGG通路富集分析。运用Auto Dock、PyMol对活性成分与关键靶点进行分子对接验证。通过药物利用度和类药性作为筛选条件筛选得到蒙药五味甘露的活性成分84种、潜在靶点773个;其中与银屑病相关的靶点基因194个。GO富集分析共获得通路674条。KEGG富集分析获得通路186条。发现蒙药五味甘露治疗银屑病是多成分、多靶点、多通路的作用结果,为深入研究蒙药五味甘露治疗银屑病的作用机制提供了理论依据。 The potential action mechanism of Mongolian medicine Wuwei Ganlu in treating psoriasis was explored,to provide theoretical basis for follow-up further experimental research.TCMSP database,Swiss ADME and Swiss Target Prediction was used to excavate the active ingredients and target genes of Mongolian medicine Wuwei Ganlu(Fringe sagebrush,Myricaria germanica,Ephedra sinica,Juniperus rigida leaves and Manchurian Rhododendron);TTD,GeneCards and other databases were used to obtain the action targets of the disease,Venn diagram was drawn according to the intersection targets of the drug and the disease,and protein interaction diagram was obtained through the intersection targets.GO function and KEGG pathway enrichment were analyzed by Metascape platform.Auto Dock and PyMol were used to verify the molecular docking between active ingredients and key targets.84 active ingredients and 773 potential targets of Mongolian medicine Wuwei Ganlu were screened using drug utilization and drug-likeness as screening criteria;Among them,there were 194 target genes related to psoriasis.GO enrichment analysis obtained a total of 674 pathways.KEGG enrichment analysis obtained 186 pathways.This study found that the treatment of psoriasis with Mongolian medicine Wuwei Ganlu was the result of multi-component,multi-target,and multi pathway action,which provided a theoretical basis for further research on the action mechanism of Mongolian medicine Wuwei Ganlu in the treatment of psoriasis.
