金线兰NPR1基因克隆及其抗性响应表达分析

摘要 【目的】克隆金线兰NPR1基因家族成员,分析其在根、茎、叶及不同植物生长调节剂处理下的表达模式。【方法】采用RT-PCR法克隆金线兰NPR1基因,进行生物信息学分析,并利用实时荧光定量PCR分析其在不同组织及不同植物生长调节剂处理下的表达。【结果】克隆得到两条长度分别为1803 bp和1401 bp的NPR1基因序列,分别命名为ArNPR1-1和ArNPR1-2。ArNPR1基因在金线兰不同部位表达具有组织特异性,其在茎中表达较低,在根、叶表达较高。ArNPR1基因对不同植物生长调节剂SA、MeJA处理的响应模式存在差异;SA处理后12 h和24 h,ArNPR1基因的表达量均上调;MeJA处理后12 h,ArNPR1基因的表达量上调,随着处理时间的延长,表达量下调。【结论】成功克隆2个金线兰NPR1基因,明晰了其在不同组织及SA、MeJA处理下的表达特征,为后续基因功能验证与应用奠定基础。 【Objective】Cloning members of the NPR1 gene family in Anoectochilus roxburghii and analyzing their expression patterns in roots,stems,and leaves,and under different plant growth regulator treatments.【Method】The NPR1 gene of Anoectochilus roxburghii was cloned using RT-PCR,and bioinformatics analysis was performed.Real time fluorescence quantitative PCR was used to analyze its expression in different tissues and under different plant growth regulator treatments.【Result】Two NPR1 gene sequences with lengths of 1803 bp and 1401 bp were cloned and named ArNPR1-1 and ArNPR1-2,respectively.The expression of ArNPR1 gene in different parts of Anoectochilus roxburghii had tissue specificity,with lower expression in the stem and higher expression in the roots and leaves.The response patterns of ArNPR1 gene to different plant growth regulators SA and MeJA treatments varied;The expression level of ArNPR1 gene was upregulated at 12 and 24 hours after SA treatment;After 12 hours of MeJA treatment,the expression level of ArNPR1 gene was upregulated,and with the prolongation of treatment time,the expression level was downregulated.【Conclusion】This experiment successfully cloned two NPR1 genes from Anoectochilus roxburghii and analyzed their expression in different tissues and under SA and MeJA treatments,providing a basis for gene function validation and utility in the future.
