山茱萸新苷介导NF-κB/NLRP3通路对脓毒症大鼠肠黏膜屏障的影响

The effect of cornus officinalis glycoside on the intestinal mucosal barrier of septic rats by regulating the NF-kB/NLRP3 pathway

摘要 目的探讨山茱萸新苷对脓毒症大鼠肠黏膜屏障的影响及可能的作用机制。方法将90只SD大鼠随机分为假手术组、模型组、地塞米松组(10 mg/kg)、山茱萸新苷低剂量组(12.5 mg/kg)、山茱萸新苷中剂量组(25 mg/kg)和山茱萸新苷高剂量组(50 mg/kg),每组15只。除假手术组外,其余各组大鼠均采用盲肠结扎穿孔术(CLP)制备脓毒症模型。尾静脉注射药物干预,术后24 h取大鼠血清及回肠组织,采用苏木精-伊红(HE)染色法观察小肠黏膜病理损伤情况;酶联免疫吸附试验(ELISA)检测血清中黏膜屏障通透性标志物肠型脂肪酸结合蛋白(I-FABP)和二胺氧化酶(DAO)水平及小肠黏膜分泌型免疫球蛋白(sIgA)水平;异硫氰酸荧光素-葡聚糖(FITC-Dextran)示踪法检测肠黏膜通透性;ELISA法检测血清中白介素-1β(IL-1β)、白介素-6(IL-6)、白介素-18(IL-18)和肿瘤坏死因子-α(TNF-α)表达水平;蛋白免疫印迹法(Western blot)检测小肠组织中核转录因子κB(NF-κB)p65、p-NF-κB p65、NOD样受体热蛋白结构域相关蛋白3(NLRP3)和含半胱氨酸的天冬氨酸蛋白水解酶1(caspase-1)蛋白表达水平。结果HE染色结果显示,与假手术组比较,模型组大鼠肠黏膜结构缺失,绒毛缩短,炎症细胞浸润;与模型组比较,山茱萸新苷中、高剂量组及地塞米松组大鼠肠黏膜结构相对完整,绒毛排列紧密,炎症细胞浸润减少,而山茱萸新苷低剂量组大鼠肠黏膜结构、绒毛排列、炎症细胞浸润等病理学改善不明显。模型组大鼠血清I-FABP、DAO水平显著高于假手术组(均P<0.05),肠黏膜sIgA水平低于假手术组(P<0.05)。山茱萸新苷中、高剂量组及地塞米松组大鼠血清I-FABP、DAO水平低于模型组(均P<0.05),肠黏膜sIgA水平高于模型组(P<0.05),而山茱萸新苷低剂量组大鼠血清I-FABP、DAO和肠黏膜sIgA水平与模型组差异无统计学意义(均P>0.05)。模型组大鼠血清FITC-Dextran含量高于假手术组(P<0.05);山茱� Objective To investigate the effect of cornus officinalis glycoside on intestinal mucosal barrier in septic rats and its possible mechanism of action.Methods A total of 90 Sprague-Dawley(SD)rats were randomly divided into a sham operation group,a model group,a dexamethasone group(10 mg/kg),a low-dose group of cornus officinalis glycoside(12.5 mg/kg),a medium dose group of cornus officinalis glycoside(25 mg/kg),and a high-dose group of cornus officinalis glycoside(50 mg/kg),with 15 rats in each group.Except for the sham surgery group,all other groups of rats were treated with cecal ligation and puncture(CLP)to prepare sepsis models.Drug intervention was administered via tail vein injection,and serum and ileal tissue were collected from rats 24 hours after surgery.Hematoxylin eosin staining(HE)was used to observe the pathological damage of the small intestinal mucosa;Enzyme linked immunosorbent assay(ELISA)was used to detect the levels of intestinal fatty acid binding protein(I-FABP)and diamine oxidase(DAO),markers of mucosal barrier permeability,as well as the levels of small intestinal mucosal secreted immunoglobulin(slgA)in serum;The FITC Dextran tracer method was used to detect intestinal mucosal permeability;ELISA method was used to detect the expression levels of interleukin-1β(IL-1β),interleukin-6(IL-6),interleukin-18(IL-18),and tumor necrosis factor-α(TNF-α)in serum;Western blot was used to detect the protein expression levels of nuclear transcription factor(NF-kB)p65,p-NF-kB p65,NOD like receptor heat protein domain associated protein 3(NLRP3),and cysteine containing aspartic protease 1(caspase-1)in small intestine tissue.Results HE staining results showed that compared with the sham surgery group,the model group rats had a loss of intestinal mucosal structure,shortened vlli,and infiltration of inflammatory cells;Compared with the model group,the intestinal mucosal structure of rats in the middle and high dose groups of cornus officinalis glycoside and dexamethasone group was relatively intact,with tig
