脱水胁迫下花生根系生理特性和基因表达变化分析

摘要 为探究脱水胁迫下花生根系生理特性和基因表达变化,本试验对花生幼苗进行脱水处理,观察其根系状态,测定其根系总长度、体积和表面积等形态指标,对根尖细胞进行染色,测定根尖ATP(Adenosine5’-triphosphate)含量和线粒体相关基因的表达量。结果表明,脱水胁迫后根系状态较为萎蔫,总长度、体积、表面积和平均直径均明显降低,根尖细胞大量死亡,根系ATP含量明显降低,线粒体相关基因表达受到干扰,可能是线粒体能量生产功能障碍,ATP合成减少,导致根系细胞死亡和根系生长受阻。研究为进一步分析脱水胁迫对花生根系线粒体功能的影响提供参考。 To investigate the physiological characteristics and gene expression changes of peanut roots under dehydration stress,the peanut seedlings were dehydrated,their root status were observed,their total root length,volume,surface area and other morphological indicators were measured,root tip cells were stained,and root tip ATP(Adenosine 5’-triphosphate)content and mitochondrial related gene expression levels were measured.The results showed that after dehydration stress,the root system was more wilted,with a significant decrease in total length,volume,surface area,and average diameter.A large number of root tip cells died,and the ATP content in the root system was significantly reduced.The expression of mitochondrial related genes were disrupted,which may be due to mitochondrial energy production dysfunction,reduced ATP synthesis,lead to root cell death and root growth inhibition.The research provided a reference for further analyzing the effects of dehydration stress on the mitochondrial function of peanut roots.
