普鲁兰多糖基pH/氧化还原响应型水凝胶的制备及性能研究

摘要 刺激响应性和可生物降解性天然聚合物基水凝胶是药物缓释载体研究的重要发展方向。基于此开发了基于普鲁兰多糖(Pullulan)的p H/氧化还原响应水凝胶。以Pullulan为基体,以甲基丙烯酸二甲氨乙酯(DMAEMA)和丙烯酸(AA)为p H响应性单体,以具有N, N'-双(丙烯酰)胱胺(BAC)为交联剂,制备具有p H/氧化还原响应性的水凝胶(PPAD),通过调控p H值和溶胀-收缩机制来控制OMT缓慢释放。载药水凝胶在p H=1.5、p H=6.8和p H=7.4时,对氧化苦参碱(OMT)的累计释放量分别为66.7%、40.0%和35.2%。PPAD1~6在p H=1.5的累计释放量分别为60.34%、66.7%、74.0%、73.0%、63.5%、54.6%,氧化还原降解实验验证了PPAD的氧化还原性。 Stimuli-responsive and biodegradable natural polymer-based hydrogels are an important development in the study of slow-release carriers for drugs.Based on this pH/redox responsive hydrogels based on Pullulan polysaccharide(Pullulan)were developed.Pullulan as matrix,dimethylaminoethyl methacrylate(DMAEMA)and acrylic acid(AA)as pH-responsive monomers,and redox-responsive BAC as cross-linking agent were used to prepare pH/redoxresponsive hydrogels(PPADs),which were used to control the slow release of OMT by modulating the pH and the swelling-contraction mechanism.The cumulative release of OMT from the drug-loaded hydrogels was 66.7%,40.0%,and 35.2%at pH=1.5,pH=6.8,and pH=7.4,respectively.the cumulative release of PPAD1 to 6 at pH=1.5 was 60.34%,66.7%,74.0%,73.0%,63.5%,and 54.6%,respectively.
