摘要
【目的】应用网络药理学方法探明青蒿抗猪繁殖与呼吸综合征病毒作用机制。【方法】用TCMSP数据库检索青蒿的可能活性成分和药物作用靶点,通过CTD数据库和GeneCards数据库收集猪繁殖与呼吸综合征病毒疾病靶点,筛选出药物和疾病靶点的交集来构建蛋白质互作网络并进行GO功能与KEGG通路富集分析,对筛选出的主要活性成分与核心靶点进行分子对接验证。【结果】青蒿有19种可能活性成分和268个药物靶点,主要可能活性成分有槲皮素、山奈酚和木犀草素。PRRSV疾病靶点有3366个,其与青蒿的交集靶点有126个。青蒿抗猪繁殖与呼吸综合征病毒的核心靶点有白介素-6、肿瘤坏死因子(TNF)、白介素-1β、前列腺素内过氧化物合成酶2、基质金属蛋白酶9、半胱氨酸天冬氨酸蛋白酶3和AP-1转录因子亚单位等。GO功能富集涉及对外源性物质刺激的反应、基因表达正调控、细胞外间隙、细胞表面、酶结合及蛋白结合等,KEGG通路主要涉及TNF、白介素-17(IL-17)等信号通路。分子对接结果显示青蒿主要活性成分与核心靶点均能实现自发结合。【结论】青蒿抗猪繁殖与呼吸综合征病毒主要有效成分可能为槲皮素、山奈酚、木犀草素,可能通过作用于IL-17和TNF信号通路发挥抗猪繁殖与呼吸综合征病毒作用。
[Objective]This study aimed to explore the mechanism of Artemisia annua(ART)against porcine reproductive and respiratory syndrome virus(PRRSV)with network pharmacology.[Methods]The possible active ingredients and drug targets of the ART were retrieved from the TCMSP database,the PRRSV disease targets were collected from the CTD database and GeneCards database,and the intersection of the drugs and disease targets was screened to construct a protein-protein interaction network,and GO function and KEGG pathway enrichment analysis were performed.Furthermore,molecular docking was conducted to validate the selected main active ingredients and core targets.[Results]The ART has 19 possible active ingredients and 268 drug targets,with the main active ingredients being quercetin,kaempferol,luteolin.There are 3366 disease targets of PRRSV,with 126 intersecting targets with the ART.The core targets of the ART against PRRSV include interleukin-6,tumor necrosis factor(TNF),interleukin-1β,prostaglandin endoperoxide synthase 2,matrix metalloproteinase 9,cysteine aspartate protease 3,and AP-1 transcription factor subunit.GO functional enrichment involves response to xenobiotic stimuli,positive regulation of gene expression,extracellular space,cell surface,enzyme binding and protein binding.The KEGG pathway mainly involves signaling pathways such as TNF and interleukin-17.The molecular docking results showed that all of the main active ingredients of the ART can spontaneously bind to the core targets.[Conclusion]The main active ingredients of the ART against PRRSV may be quercetin,kaempferol and luteolin,which may exert anti-PRRSV effects by acting on the TNF and IL-17 signaling pathways.
