摘要
目的探讨桂枝加龙骨牡蛎汤改善魄不安于肺型不寐大鼠学习记忆的作用机制。方法将48只SD雄性大鼠按照随机数字法分为造模组(36只)和正常组(12只)。造模组采用多组不稳定平台浅水环境法部分剥夺睡眠42 d。在第15 d随机分为模型组、右佐匹克隆组(西药组)、桂枝加龙骨牡蛎汤组(中药组),每组12只,再相应干预28 d。以水迷宫实验评价模型大鼠学习记忆功能及干预后的疗效,以脑电监测评价模型大鼠的快速动眼睡眠时间(REM)、非快速动眼睡眠时间(NREM)、觉醒次数、睡眠片段平均时间及各干预组的疗效。以尼氏染色观察大鼠海马齿状回多形层神经元的变化,以免疫组化法检测肿瘤坏死因子α(TNF-α)、白细胞介素1β(IL-1β)及IL-6的变化。结果与正常组比较,模型组第2、3、4、5 d逃避潜伏期显著延长(P<0.05),穿越平台总次数显著减少(P<0.05),NREM、REM、平均睡眠片段长度显著缩短(P<0.05),觉醒次数显著增加(P<0.05),神经元尼氏体数量变少,胶质细胞增多,脑TNF-α、IL-1β、IL-6表达水平均显著升高(P<0.05)。与模型组比较,西药组第3 d和中药组第2、3、4、5 d逃避潜伏期均缩短(P<0.05),西药组和中药组穿越平台总次数均增加(P<0.05),NREM、REM、平均睡眠片段时间均显著增加(P<0.05),觉醒次数均显著降低(P<0.05),尼氏体增多,胶质细胞下降,脑TNF-α、IL-1β、IL-6相对表达水平均显著降低(P<0.05)。与西药组比较,中药组虽然逃避潜伏期均值较低,但无统计学差异(P>0.05),穿越平台总次数显著增加(P<0.05),REM、觉醒次数显著增加(P<0.05),平均睡眠片段长度显著下降(P<0.05),海马神经元较多,胶质细胞明显减少,脑TNF-α、IL-1β、IL-6显著下降(P<0.05)。结论桂枝加龙骨牡蛎汤能够显著改善魄不安于肺型不寐大鼠的睡眠及学习记忆功能,其部分机制可能与下调脑TNF-α、IL-1β和IL-6等炎症因子的水平有关。
Objective To investigate the effects of GuizhiLongu Muli Decoction(GLMD)on memory function of model insomnia rats with Pounrest in the lung based on some inflammatory factors in the brain.Methods Forty-eight Sprague-Dawley(SD)male rats were randomly distributed into the normal group(n=12)and model preparing group(n=36).The rats in the model preparing group weredeprived of partial sleep using the unstable multiple platforms surrounded by shallow waterfor 42 days.On day 15,the 36 model preparing rats were randomly divided further into model group,eszopiclone group and GLMD group and intervened for 28 days(n=12).Morris Water Maze test was used to evaluate the learning and memory function as well as the memory improvement of the model rats.Electro encephalo gram(EEG)monitoring was used to evaluate time of rapid eye movement(REM)and non-REM(NREM),arousaltimes,average sleep bout,as well as the efficacy of intervention group.Nissl staining was used to observe the changes of multiform neurons in the dentate gyrus of the rat hippocampus,and the immunohistochemistry was used to detect changes intumor necrosis factor-alpha(TNF-α),interleukin-1 beta(IL-1β),interleukin-6(IL-6).Results Compared with the normal group,on days 2,3,4,and 5,the escape latency of the model group was significantly prolonged,the number of platform crossing was significantly reduced,the time length of NREM,REM and average sleep bout was significantly shortened(all P<0.05),the arousal times was significantly increased(P<0.05),the number of Nissl bodies in neurons decreased,glial cells increased,TNF-α,IL-1βand IL-6 increased significantly(P<0.05).Compared with the model group,the escape latency on day3 in the eszopiclone group and which on days 2,3,4,and 5 in GLMD group were significantly shorter(P>0.05),the number of platform crossing increased slightly in eszopiclone group and GLMD group(P<0.05),and the time length of NREM,REM and average sleep bout significantly prolonged,the arousal times significantly decreased(all P<0.05),Nissl bodies increased
