摘要
目的探究茯苓酸调节Hippo-Yes相关蛋白(YAP)信号通路对多囊卵巢综合征(PCOS)大鼠的保护作用。方法将大鼠分为空白组、模型组、二甲双胍组(100 mg/kg)、茯苓酸低(10 mg/kg)、高剂量组(40 mg/kg)、PY-60组(YAP激活剂PY-6010μmol/L)、茯苓酸高剂量+PY-60组(40 mg/kg茯苓酸+10μmol/L PY-60),每组10只。给予相应的处理后,检测大鼠空腹血糖(FBG)、空腹胰岛素(FINS)水平;观察卵巢组织病理损伤和细胞凋亡情况;检测大鼠血清卵泡刺激素(FSH)、黄体生成素(LH)、睾酮(T)和雌二醇(E2)水平;分析大鼠卵巢组织中Hippo-YAP信号通路相关蛋白表达。结果与空白组相比,模型组大鼠动情间期天数、体重、FBG、FINS、胰岛素抵抗指数(HOMA-IR)、细胞凋亡率、囊性卵泡数、T、LH/FSH和LH水平及磷酸化(p)-YAP、p-PDZ结合基序转录共激活因子(TAZ)蛋白水平升高,E2、FSH、黄体数降低(P<0.05);与模型组相比,茯苓酸低、高剂量组、二甲双胍组大鼠相应指标变化趋势与上述相反。PY-60减轻了茯苓酸改善PCOS大鼠卵巢损伤的作用。结论茯苓酸可能通过抑制Hippo-YAP通路改善PCOS大鼠卵巢损伤。
Objective To investigate the protective effect of poric acid in polycystic ovary syndrome(PCOS)rats by regulating the Hippo-Yes associated protein(YAP)signaling pathway.Methods Rats were divided into blank group,model group,metformin group(100 mg/kg),poric acid low(10 mg/kg),high dose group(40 mg/kg),PY-60 group(YAP activator PY-6010μmol/L),and high-dose poric acid+PY-60 group(40 mg/kg poric acid+10μmol/L PY-60),with 10 rats in each group.After treatment,the body weight,FBG,and FINS levels of rats were detected,and the HOMA-IR was calculated.The pathological changes and apoptosis of ovarian tissue were observed;serum levels of FSH,LH,T,and E2 were detected;Western blot was used to detect Hippo-YAP signaling pathway-related protein expression in rat ovarian tissue.Results Compared with the blank group,the number of days between estrus,body weight,FBG,FINS,homeostatic model assessment-insulin resistance(HOMA-IR),cell apoptosis rate,number of cystic follicles,the levels of T,LH/FSH,LH,and the protein expression of phosphorylated(p)-YAP,p-the transcriptional co-activator with PDZ-binding motif(TAZ)in the model group were obviously increased,the E2,FSH,and corpus luteum count were obviously decreased(P<0.05).Compared with the model group,the change trend of corresponding indexes in the low dose,high dose and metformin groups was opposite to the above.PY-60 alleviated the effect of poric acid in improving ovarian damage in PCOS rats.Conclusion Poric acid may improve ovarian damage in PCOS rats by inhibiting the Hippo-YAP pathway.
