参连肠炎丸中4种活性成分含量测定

摘要 目的利用高效液相色谱切换波长的方法同时测定参连肠炎丸中甘草苷、延胡索乙素、橙皮苷和盐酸小檗碱含量。方法色谱柱为Agilent Eclipse XDB-C_(18)(5μm,4.6 mm×250 mm),流动相为乙腈-0.2%磷酸,进行梯度洗脱,流速为1.0 ml/min,甘草苷、延胡索乙素、橙皮苷和盐酸小檗碱的检测波长和切换时间分别为237 nm(0~7 min)、283 nm(7~12 min)、283 nm(7~12 min)、345 nm(12~20 min)。结果甘草苷、延胡索乙素、橙皮苷和盐酸小檗碱4种成分良好分离,分离度均在1.5以上。4种成分线性关系良好,平均回收率分别为100.19%,99.32%,99.64%,100.08%,RSD分别为1.78%,1.69%,1.89%,2.04%(n=6)。结论本法操作简单、结果准确,回收率高,重复性好,可为参连肠炎丸质量的有效控制提供一定的技术支撑。 Objective To simultaneously determine the contents of glycyrrhizin,tetrahydropalmatine,hesperidin and berberine hydrochloride in Shenlian Changyan Pills by HPLC-wavelength switching method.Methods The chromatographic column was Agilent Eclipse XDB-C_(18)(4.6 mm×250 mm,5μm)column with acetonitrile-0.2%phosphoric acid solution as mobile phase for gradient elution.The flow rate was 1.0 ml/min.The detection wavelengths and switching time of glycyrrhizin,tetrahydropalmatine,hesperidin and berberine hydrochloride were 237 nm(0-7 min),283 nm(7-12 min),283 nm(7-12 min)and 345 nm(12-20 min),respectively.Results Glycyrrhizin,tetrahydropalmatine,hesperidin and berberine hydrochloride could be well separated,and the resolutions were all above 1.5.The four components had good linear relationships.The average recoveries of the four components were 100.19%,99.32%,99.64%and 100.08%,respectively,and the RSDs were 1.78%,1.69%,1.89%and 2.04%(n=6),respectively.Conclusion This method has the advantages of simple operation,accurate results,high recovery and good repeatability.It can provide some technical support for the effective quality control of Shenlian Changyan Pills.
